Welcome to 181LSection 14 Room 430

This lab is about you:Thinking, Evaluating, Investigating and Understanding

This lab is NOT about you:Showing up, jumping through hoops and

leaving

We are not synchronized with the lectures, but cover ‘same’ material

Welcome to 181L

• Read the lab manual BEFORE lab• Be ON TIME to class• PARTICIPATE• Complete the assignments (online, etc)• Participate• Think, evaluate, investigate, understand

Expectations

• Phones, etc• Food and drinks• C’mon, really?• Code of Conduct http://deanofstudents.arizona.edu/studentcodeofconduct

Expectations

• Info found in syllabus• Assignments• Online assessments/tutorials 10%• In-class quizzes 15%• Lab-based assignments 50%• Projects and Reports 25%

Grading

• Absences must be excused through Asya Roberts in BSE 109• Arriving late/leaving early is absent• Make-up labs are labs. Only current week• Miss 2 labs and you should drop, or we’ll do it

Attendance

• Just do YOUR work• Plagiarism agreement p xii• Link http://deanofstudents.arizona.edu/codeofacademicintegrity/

Molecules and the Tao of the Molecular World

Molecules and the Tao of the Molecular World

Molecules and the Tao of the Molecular World

Molecules and the Tao of the Molecular World

How molecules feel, and the world they live in

Molecules and the Tao of the Molecular World

• Observe the forces acting on and between macromolecules

Molecules and the Tao of the Molecular World

• Observe the forces acting on and between macromolecules • Make deductions about molecular world from observations

Molecules and the Tao of the Molecular World

• Observe the forces acting on and between macromolecules • Make deductions about molecular world from observations• Become familiar with computer representations of molecules

Molecules and the Tao of the Molecular World

• Observe the forces acting on and between macromolecules • Make deductions about molecular world from observations• Become familiar with computer representations of molecules• Begin to think of biological molecules as real, with distinctive surfaces.

Atoms: They’re how life works

DNA: C, H, N, O, P, [Mg++]RNA: C, H, N, O, P, [Mg++]Protein: C, H, N, O, (S), (P), [traces]Membranes: C, H, N, O, P, (S)

It’s pretty simple, really…

What cannot be done with assemblages of these atoms* cannot be done by living

organisms, nor their cells, nor their spit, etc.

Who Am I?• At birth, # protons = # electrons• Atoms seek completion• Mostly it means 8 in the outer ‘shell’• Hydrogen and Helium are two

ColorsYou’ll need

to know thesefor NEXT week

ColorsRED also means ‘negative’

Blue also means ‘positive’

A Little JargonBonds/Interactions

Covalent bond: shared pair of electrons* Non-polar bond: equal sharing; “greasy” Polar bond: doesn’t share well with others Hydrogen bond: Hydrogen interaction-- partial(+) H atom attracted to partial(-) oxygen or nitrogen Ionic bond: attraction between opposites

Hydrophobicity (non-polar covalent bonds)

Water is disordered - when lipids present, water molecules arrange in more ordered way.

Clumping together of fat molecules reduces amount of water that has to be ordered.www.steve.gb.com/science/membranes.html

Alcohol vs. Water• “If this is true…”• Densities:

• Which one is closest to water?• Oil/Water and/or Salt Water/water

MeOH = 791EtOH = 789PrOH = 803BuOH = 810

Ethanol vs. Water

•Which is bigger/heavier?•Which evaporates/distills first?•Why...?

Clean Up• Oil waste in the hood• Ethanol waste in sink• Anything too messy, throw away• Leave the lab as you found it
