Process Overview
TFF/Cross Flow Filtration Process Overview Atangentialflowfiltration(TFF)orcrossflowfiltrationprocessusingahollowfiberfiltermodule(versusaplate&framecassetteflatsheetdevice)isrepresentedinFigure1.
Liquidisfedfromaproductvesselbyacirculationpumptoafiltermodulecontainingnumeroushollowfibersandtheliquidflowsintotheinnercrosssectionoftheindividualfibers.
Thewallofthehollowfiberisthefiltermembrane.AsisshowninFigure2,acertainamountofliquidandliquidcomponentssmallerthantheporesizeofthefiberwall“permeates”throughthewall(thefiltrate).Thefiltratecollectsinthemoduleshellandtheshellhasportsforremovaloffiltratefromtheshell(thevoidareabetweenthefibersisclosedatbothendstopreventtheliquidthatisenteringthefiltermodulefromgoingdirectlyintotheshell).Thefeedpressure(PIN)ishigherthantheretentatepressurein(POUT)becauseofthepressuredropasliquidflowsthroughthenarrowfibersandreturnstothevessel.ThereisalsoapressuredropacrossthefiberwallandthefiltratepressureismeasuredatPF.Monitoringofthesepressuresiscriticaltomeasure processperformanceandforprocesscontrol.Thecrossflowrateisordersof magnitudeabovethefiltrateflowrateanditisthisphenomenathatpreventsamembranefilterfromcloggingfrommaterialthatwouldrapidlyclogamembranefilteroperatingin“normalflow”filtration.Inflatsheetdevices,eventhoughthegeometryofthedeviceisdifferent,theoperationoftheprocessissimilar.TFFisaveryeffectiveprocessselectioninmanyareasofbiopharmaceuticalprocessing.Dependingontheprocess,afiltratepumpmaybeusedtolimitthefiltrateflowandpreventrapidfilterfouling.Athrottlingvalvemaybeusedontheretentatetubetocreatebackpressuretodriveliquidthroughthemembrane.InaTFFprocess,asfiltrateisremoved,thevesselcontentsareconcentrated.Inadiafiltrationprocess,liquidisaddedtotheproduct vesselasthesamerateoffiltrateremoval.

Thesystemcomeswiththerequiredcablestoenablethesytemtobequicklyupandrunning.Allconnectionsarekeyedtopreventconnectionofacabletothewrongconnector.Pumpsandscalesmaybedeliveredwiththesystemorexistingequipmentorself-procuredequipmentmaybeused.
Thevalvereceivesthepressuresignalfrom the control system and controls backpressurebyvaryingtheflowpatharea by only contacting the outside of thetubingwithoutthevalvecontactingthefluid.Itincludesone-touchreal-timeadjustment of the proportional setting for thePIDcontroltoaccountfordifferentpumps and system setups.

Connecting to this input, integrates the reading into the option Retentate Flow Control algorithm.

Thenon-invasiveultrasonicairdetectordetectsairinatubeplacedintothedetector.ThereisanintegralLEDthatindicatesliquidpresence.Thetubecanbeopaquebecausethesensingmechanismissoundwaves.Thesoftwarehasauserentereddetectiondelaytopreventfalseendpoints.
TheAirDetectorwhichdetectsthedifferencebetweenairandliquidinatube,isintegratedintothesoftwareasaDiafiltrationEndpoint.Thiscanbeusedprimarilyinoneoftwoways.Oneistorunafed-batchprocesswhereliquidiscontinuouslyfedtoallowabatchtobecontinuouslyaddedtothevesselwhichsimultaneouslyconcentrates.Thisgivesmoreflexibilitytoadefinedsetupandcanassistinminimizationoftheoverall systemhold-upbyminimizingthevesselsize.Secondly,itcanbeforusedwithapre-measuredbuffervolumeforadiafiltrationstep.
