About the role: An opportunity will be available from April 2018 within the Early Detection theme of the CRUK Lung Cancer Centre of Excellence (CoE) at CRUK MI, Manchester and University College London. The successful candidate will initially spend up to 6 months in the biomarker laboratories of Professor Caroline Dive CBE at the CRUK Manchester Institute linking to Dr Philip Crosbie’s respiratory health clinical research at the University Hospital of South Manchester. They will then be hosted in Professor Avrum Spira’s laboratory at Boston University Medical Center in Boston, USA for a period of up to 2.5 years, returning to CRUK MI, Manchester to complete their research. The position is designed to build upon strong collaborations between the CRUK Lung CoE and Prof Spira’s laboratory and to develop the post holder’s career such that they can subsequently apply for a competitive, independent CRUK funded fellowship in the early detection of lung cancer within our CRUK Lung Cancer CoE. JOB PURPOSE To gain a thorough scientific training in the early detection of lung cancer and/or biology of early lung cancers. To conduct high quality research overseen by the leaders of the Early Detection/Early Disease themes of the CRUK Lung Cancer Centre of Excellence and by Professor Spira. To agree, develop and deliver a defined and independent research project that complements the research of the Centre and strengthens the existing collaboration with Prof Spira’s laboratory. The post holder will initially spend up to 6 months in the biomarker laboratories of Professor Caroline Dive CBE at the CRUK Manchester Institute linking to Dr Philip Crosbie’s respiratory health clinical research at the University Hospital of South Manchester. They will then be hosted in Professor Avrum Spira’s laboratory at Boston University Medical Center in Boston, USA for a period of up to 2.5 years, returning to CRUK MI, Manchester to complete their research. It is anticipated that the post holder will, by the end of this post, be in a competitive position to establish an independent research program focused on early lung cancer.

BACKGROUND Lung cancer is the most common cause of cancer related death in the world; more than 1.6 million people die of the disease each year. Late clinical presentation is common, with approximately 75% of patients having either locally advanced (stage III) or distant metastatic disease (stage IV) at diagnosis, when treatments are much less effective and survival poor. In the UK ≈100 patients die

from lung cancer every day and only 5% survive 10 years. In Manchester, lung cancer is the single biggest cause of premature death, reflecting high levels of smoking and socio-economic deprivation. Understanding the biology of early stage disease is a critically important component of our strategy to improve outcomes for patients with lung cancer, as this is the area where the greatest impact can be made. The CRUK Lung Cancer of Excellence led by Professor Caroline Dive and Professor Charles Swanton, located in sister sites at Manchester University and University College London, is composed of eight integrated themes including Early Disease and Early Detection, Biomarkers, Basic Research, Tumour Heterogeneity, Immunology, Bioinformatics and Advanced Radiotherapy and Radiobiology and Clinical Trials. Working within the CRUK MI CEP group, led by Professor Dive, this fellowship post will develop a new programme of research in early stage lung cancer biology and/or early detection of lung cancer. The post holder will be able to interact with members of the early disease and early detection theme, led in our Lung Centre by Dr Philip Crosbie at the Manchester University NHS Foundation Trust (MFT) and at UCL by Professor Sam Janes. Early detection of cancer and lung cancer are also strategic foci of the CRUK Manchester Centre. The post holder will benefit from the strong local research environment in Manchester. The CEP group (http://www.cruk.manchester.ac.uk/Research/CEP) and newly-established Manchester Centre for Cancer Biomarker Sciences, led by Professor Dive, will host this fellowship. The group has an international reputation for liquid biopsies, specifically in advanced lung cancer, including circulating tumour cells (CTCs) and circulating DNA (Girotti et al, Cancer Disc. 6(3):286-99, 2016, Gremel et al, Ann Oncol. 1959-65. doi: 10.1093/annonc/mdw278, 2016) and circulating miRNAs with Dr Michela Garofalo at CRUK MI. The CEP group pioneered unique patient CTC derived explant models in mice (CDX) (Hodgkinson et al, Nat Med. 23(1):114-119, 2017) and is developing panels of lung cancer CDX and PDX, together with orthotopic models. They focus predominantly on small cell lung cancer. The CEP group works closely with Thoracic Medical Oncologist Professor Fiona Blackhall at the adjacent The Christie NHS Foundation Trust. The post holder will be strongly encourage to interface with clinical colleagues at MFT, who provide a comprehensive multi-disciplinary thoracic oncology service to the local population and wider Manchester Cancer Network (n=2,200 new lung cancer diagnoses per year). Specialist lung cancer services at MFT include: a RAPID lung cancer diagnostic pathway, standard/advanced bronchoscopy (>2,000 procedures per year) e.g. endobronchial ultrasound (EBUS), based in the UK’s largest purpose built bronchoscopy unit (The Lucis Centre), interventional thoracic radiology services, a survivorship programme and a pleural team. Medical and clinical oncologists deliver adjuvant chemotherapy and treatments for advanced lung cancer and mesothelioma. MFT hosts one of the country’s largest thoracic surgical units, with five dedicated thoracic surgeons who perform >450 primary lung cancer resections a year. Access to lung and tumour tissue for research is facilitated through a well-established collaboration with the Manchester Cancer Research Centre biobank. The service and biobank are supported by a specialist thoracic pathology team. Lung cancer research at MFT is co-ordinated by the Thoracic Oncology Research Hub (TORCH), a collaboration of clinically active investigators that covers the entire patient journey. The Lung Cancer Team at MFT is an academic department with a substantial early detection research portfolio. Screening studies, led by Dr Philip Crosbie, ongoing or upcoming include: 1. The Manchester Early Detection of Lung Disease (MEDLD) Pilot (funded by Macmillan Cancer Support): a community-based lung cancer screening pilot using low dose CT (LDCT), targeted at high-risk (hard-to-reach) smokers in deprived areas of Manchester (n=2,541). 2. The Community Lung Health Study (CLHS) (funded by Cancer Research UK): This study is a collaboration with CEP and the Manchester Collaborative Centre for Inflammation Research (MCCIR)

with concomitant collection of blood, sputum and nasal samples for discovery of minimally invasive early detection biomarkers. Samples have been collected on ≈600 individuals at high risk of lung cancer (2nd round attendees of the MEDLD pilot). 3. The North Manchester Lung Health Study (NMLHS) (funded by Everest Detection): this study will start in Q3/4 2018 and will screen high risk individuals using annual low dose CT (LDCT) over 2 screening rounds (annual); bloods will be taken from all attendees (n≈5,500) for early detection biomarkers. 4. The Yorkshire Lung Screening Trial (YLST) (funded by Yorkshire Cancer Research): will screen high risk individuals will LDCT over 2 screening rounds (biennial). It is hoped that a biomarker sub-study will run alongside the main study (n≈7,000). Collaboration with world-leading researchers of COPD is also welcomed. Professor Jørgen Vestbo leads the North West Lung Centre (NWLC) at MFT, the largest clinical respiratory department in the UK and an internationally recognised research unit with world leading research in COPD, asthma/allergy, cough and fungal infections. The NWLC includes the Respiratory Theme of the recently awarded NIHR funded Manchester Biomedical Research Centre, which has for years had a dedicated Respiratory and Allergy Clinical Research Facility, and the NWLC is closely linked with the UK’s largest commercial respiratory clinical trials facility, the Medicines Evaluation Unit. In COPD, the academic work focuses on early COPD, exacerbations, biomarkers and, pertinent to this new fellowship, links to lung cancer. The fellow will have the opportunity to interface with the ongoing early detection research within the MCCIR, led by Professor Tracey Hussell (www.mccir.manchester.ac.uk), a unique venture designed to establish a world-leading translational research centre in the UK for inflammatory diseases which affect the joints, lungs, skin, and many other organs in the body. By bringing together the best industrial and academic scientists, the MCCIR enhances our understanding of inflammatory mechanisms and develops ways in which this knowledge can be translated into the identification and clinical development of novel and improved therapies. The fellow will have access to state-of-the-art facilities including 18 colour flow cytometry, CyTOF and the wealth of expertise encompassed in 14 internationally recognised research groupings. Of particular relevance to this position is the interest in novel biomarkers of cleaved receptors and cellular matrix fragments that enable the identification of patients prone to aberrant inflammatory responses, the potential for development of therapy toxicities and more sensitive methods of detecting early disease. Additional opportunities for collaboration will be with Professor Tony Whetton, who leads the Stoller Biomarker Discovery Centre. This is the leading clinical proteomics centre in Europe with the ability to undertake discovery, validation and verification of protein biomarkers in a GCP environment. Work to date in the Centre has identified markers of risk for ovarian cancer and also potential lung cancer biomarkers using discovery proteomics plus mass spectrometry/ELISA validation techniques. Embedded in the Stoller Centre is an MRC funded Pathology Innovation Centre that is charged with developing new tests for clinical usage from findings in the Stoller Centre and elsewhere. Overall the research environment in Manchester provides a wonderful opportunity for the appointed fellow to develop and progress an exciting research programme on early stage lung cancer and/or its earlier detection. Collaborative research within the CRUK Lung Cancer Centre of Excellence At UCL, Professor Sam Janes leads the Early Disease and Early Detection theme within the Lung Cancer Centre, and the post holder will be able to interact and collaborate with the UCL team. Janes’

research focus is on lung cancer pathogenesis and the development of novel therapies to treat both early and late stage disease. Janes leads the Airway Regeneration and Carcinogenesis Group, with an interest in the mechanisms controlling cell fate decisions and defining whether it is failure of control of these cells that causes early lung cancer lesions. The group’s clinical interest is in monitoring a large cohort of patients with pre-invasive lung cancer lesions. International Collaboration with Professor Avrum Spira Dr. Spira research program at Boston University Medical Campus focuses on the translational study of genomic alterations associated with smoking-related lung disease. His research is based on the paradigm that smoking and other inhaled carcinogens create a ‘field of molecular injury’ in epithelial cells that line the respiratory tract. By sampling these relatively accessible airway cells, Dr. Spira’s lab has developed novel diagnostic and therapeutic biomarkers for lung cancer and COPD as well as a non-invasive approach for measuring the physiological response to a number of inhaled environmental exposures. He leads the DECAMP consortium which is a multicenter study within military and veteran hospitals in the US for developing and validation early detection biomarkers for lung cancer. He also leads a Stand Up To Cancer Dream Team in collaborations with Drs. Swanton and Janes focused on developing new approaches lung cancer interception.

DUTIES AND RESPONSBILITIES The Travelling Fellowship holder will agree and develop a programme of research in early stage lung cancer biology and/or early detection of lung cancer that forms a strong collaborative effort between the CRUK Lung Cancer Centre of Excellence Early Disease & Early detection Theme and Prof Avrum Spira’s laboratory. Key duties of the role include: Research

Planning, performing analysing, interpreting and presenting research studies.

Ensure appropriate experimental design and assist with troubleshooting.

Check data analysis and ensure correct statistical analysis has been performed.

Check scientific rational, study design and for all research protocols to be performed with approval from the appropriate laboratory lead.

Ensure all of the above is in accordance with institute policies. Budget management

Take responsibility for checking budget transactions. Regulatory compliance as appropriate

Take responsibility for compliance with local regulations regarding any in vivo work.

THE CITY OF MANCHESTER AND THE REGION Manchester is one of the great European cities and the Cancer Research UK Manchester Institute is located a short distance from the city centre and is serviced by regular public transport to the city centre. The city’s architecture represents one of the high points of Victorian achievement. The modern city is a major centre of banking, commerce and manufacturing. It has a highly cosmopolitan atmosphere and its cultural life is internationally renowned. There are three outstanding professional theatre companies, the halls of the Hallé and BBC Philharmonic orchestras, the Cornerhouse as well as other cinemas, and Europe’s fastest-growing Chinatown. On the way into the city, there is perhaps the best stretch of road in the entire country for Indian and Pakistani food, known as the curry mile in Rusholme. Amongst developments enriching the area’s cultural life have been: the opening of the Lowry Centre and Media City at Salford Quays; the opening of the Bridgewater Concert Hall; the refurbishment of the City Art Gallery; the opening of Urbis in the centre of Manchester and of the Imperial War Museum North, designed by Daniel Libeskind, in Trafford. Trafford, specifically Old Trafford, is known internationally for sport: it is a venue for Test cricket and the home of Manchester United FC. The Commonwealth Games were held in Manchester in 2002 and were highly acclaimed. The Commonwealth Stadium became the home of Manchester City FC in 2003. Housing is varied, plentiful and the price ranges can start moderately priced and are as varied as the requirements. Schooling ranges from world-famous private schools to excellent sixth-form colleges and comprehensives. Manchester is well served by a major international airport, with direct scheduled flights to many destinations in Europe as well as North America and Asia. Manchester Piccadilly railway station has been refurbished and is served by inter-city and other train services – with a direct link to Manchester Airport. The expanding network of Metrolink tram services offers an alternative mode of public transport from certain parts of the conurbation and is currently being expanded to include connections near to the Institute. Some of the most beautiful countryside in Europe is just short of an hour’s drive from the Institute in the Peak District National Park, while the Lake District and Snowdonia are also within easy reach. Manchester offers extensive provision for research. Library facilities include the John Rylands University Library (the major library in the North West and the third largest in the country) and the Manchester Central Reference Library. Facilities for quantitative analysis are provided by Manchester Computing and the Computer Support Unit

POSTDOCTORAL SCIENTIST TERMS AND CONDITIONS The following is a basic summary of the standard terms and conditions applicable to the post you have applied for:

The Postdoctoral Scientist grade is on a Cancer Research UK salary. Starting in the range of £30,000 – £39,800 per annum, dependent upon experience and qualifications.

Your employment will be with The University of Manchester appointed under The Cancer Research UK Manchester Institute terms and conditions.

The position will be located at CRUK Manchester Institute (CRUK MI), UK and Boston University Medical Centre, USA.

Subsistence allowance whilst based in the USA.

Salary is paid monthly.

There are 32 days holiday per year plus Bank Holidays for England.

The duration of the contract is four years.

37 hour week.

There is a probationary period attached to this post of 8 months. It’s standard for many organisations now and consists of two 4-monthly reviews with your line manager.

You are eligible to join the USS pension scheme.

Any offer made by the Cancer Research UK Manchester Institute would require the successful candidate to undergo a medical clearance. This is arranged with Occupational Health department at the University of Manchester prior to starting employment and consists of a basic medical. This is to address and gain clearance for any potential hazards identified for the role on offer.

Offer is subject to receipt of 3 satisfactory references and proof of your highest qualification.

The successful candidate is required to have submitted their thesis prior to their start date. Confirmation from their educational institution will be required by the Institute’s Human Resources Department prior to commencing employment.
