THE POWER OF INVISIBLE LIGHT: A NEW GENERATION OF MULTIBEAM OPTICAL TWEEZERSThe mmi CellManipulator features an intensely focused laser beam to hold, move, rotate, join, separate and stretch up to ten microscopic objects simultaneously.

A The mmi CellManipulator is an optical trap that enables precise, contact free mani -pulation of microscopic particles. It is capable of manipulating micrometer-sized dielec-tric particles, living cells, or subcellular organisms by exerting pico Newton forces via a highly focused laser beam. The beam is focused by sending it through a microscope objective. The narrowest point of the focused beam, known as the beam waist, contains a very strong light gradient. Dielectric particles are attracted along the gradient to the region of brightest light, the center of the beam. Using an infrared laser an invisible optical trap is created.

The mmi CellManipulator traps particles using a high-quality, Nd:Yag infrared laser with a wavelength of 1064nm and 3W maximum power.

Specially designed optics focuses the laser beam through an additional port without impairing any functionality of the inverted microscope. In addition, simultaneous use of phase contrast, DIC, fluorescence imaging, etc., together with trapping and force measurement is possible.

MOLECULAR MOTOR STUDIES:Josh Baker, Reno, Nevada, Biochemistry and Molecular Biology“Our current focus is on studying cooperative interactions among myosin motors in cells. Interestingly, myosin molecules function as force sensors as well as force gen-erators, thus when many myosin molecules function together in ensemble systems like muscle, the force generated by one myosin can cooperatively affect the biochemistry of neighboring myosin molecules. To better understand the interplay between myosin’s force generating and force sensing biochemistry, we use state-of-the-art experimen-tal techniques (laser traps, fluorescence microscopy, and single molecule imaging) to measure and systematically perturb (chemically, genetically, and biologically) the mechanics and chemistry of both single myosin motors and collective motor systems. We integrate these results into mathematical and computer models in an effort to better understand the molecular basis for cell motile processes ranging from muscle contrac-tion to intracellular transport.”

Our setup allows simultaneous optical trapping and confocal imaging.”

TECHNOLOGYThe mmi CellManipulator’s versatility combines a 3W infrared laser, 2 fast gal va-no me tric mirrors, a high precision xy-stage, and a digital mmi CellCamera, with most scienti fic inverted mircroscopes.

C The advanced mmi CellManipulator maintains and controls up to 10 separate beams, each capable of manipulating one particle through the whole field of view. The beams are generated from one laser source by utilizing the high switching speeds of the galvanometric mirrors. In addition, any number of beams can be grouped for simul-taneous manipulation such as movement, rotation or stretching.

The optional installation of the quadrant detector based positioning measurement sys-tem enables precise nanometer scale position detection resulting in pico Newton scale force measurements. Moreover, the mmi CellManipulator system can be used on its own or in combination with other components, such as the mmi CellCut Plus microdis-section system, to create a unique microdissection and micromanipulation instrument for highly advanced research.

With multiple ports and dual-level laser integration, the microscope offers „optical bench flexibility“, enabling the seamless use of different modules and imaging technologies. The system-integrated digital camera is connected to the standard left side port; how-ever, other cameras may be added at different ports according to individual needs.

mmi CELLTOOLS SOFTWARED The optical tweezer system is controlled by the user-friendly mmi CellTools soft-ware, which provides an easy-to-use interface for simple creation, organization, and manipulation of traps. The software tools provide the advanced, function of moving up to 10 independent laser beams according to your unique research needs. Additional advantages are live video presentations, recordings with sample scanning, and docu-mentation through screenshots.
