    BRIEF MANUFACTURING PROCESS

    1. CANDESARTAN CILEXETIL

    Manufacturing Process

    Stage – I:

    Methyl 2-[({2’-cyanobiphenyl-4-yl} methyl) amino]-3-nitrobenzoate reacts with stannous

    chloride in presence of ethyl acetate then reaction mass obtained by hydrolysis.

    Stage – II:

    Charge stage-I in toluene reacts with Tetra ethyl ortho carbonate in presence of acetic acid.

    Stage – III:

    Sodium azide reacts with Tri n butyl tin chloride in o-Xylene then separation and hydrolysis.

    Charge in acetone reacts with Triethyl amine the centrifugation of reaction mass. Charge in

    Dimethyl Formamide reacts with Potassium carbonate & Cyclohexyl 1-chloro ethyl

    mixture of Acetone and D M Water to get for purification for Candesartan Cilexetil pure.

    2. CELECOXIB

    Manufacturing Process

    Stage-I

    Charging of trifluoro acetic acid, cooling to 0-5°C, addition of absolute alcohol & sulphuric

    acid followed by heating of reaction mixture to 65-70°C and then after

    Stage-II

    Charging of Toluene followed by charging of sodium methoxide then cooling of mixture upto

    0-5°C then charging of ethyl trifluoro acetate(Stage-I).After completion of reaction pH

    adjustment of reaction mixture by addition of 20% sulphuric acid solution, charging of DM

    Water to the reaction mixture and then layer separation. Product extraction from aq. layer

    by Toluene. Finally isolation of Stage-II by combining of organic layer and distill out Toluene.

    Packing of Residue (Stage-II) after distillation of Toluene.

    Charging of 12.5 % Aqueous Isopropyl alcohol followed by 4-Sulfamoyl phenyl hydrazine

    hydrochloride and Stage-II Heating of reaction mixture up to 80-85°C.thus by reaction

    between 4-Sulfamoyl phenyl hydrazine hydrochloride and Stage-II at 80-85°C in presence of

    12.5% Aq. Isopropyl alcohol we obtain Crude Celecoxib (Stage-III). Charcoalization of

    reaction mixture and filtration through Hyflo bed at 80-85°C,Charging of DM Water into the

    filtrate followed by chilling of reaction mixture up to 0-5°C.Finally centrifugation , drying &

    packing of the material .

    Charging of 30% Aqueous Isopropyl alcohol followed by stage-III, dissolution at 80-85°C,

    Charcoalization of the material. Filtration through hyflo bed in hot condition, chilling of

    reaction mass to 0-5°C, centrifugation of the material. Finally material is to be dried under

    vacuum followed by milling, sifting & finally packing of the material.
