The complementation assay, as described, above, is also being used to purify the dna B gene product. Partially purified fractions, isolated from dna B ts cells, are thermolabile compared to fractions isolated from dna C ts cells. Also by this complementation assay, activities that stimulate dna C and dna D receptor crude extracts have been purified. Thus, by complementation in the sX174 DNA-dependent system, it should be possible to purify the products of all the known dna genes. Because a defined DNA template is used, it should be possible to analyze the mechanism of DNA synthesis carried out by this system.
