    Possible Modes of Action

    Vegeceuticals are standardized to Beta Carotene (Orange) and Anthocyanins (Red).

    Beta Carotene

    Regular consumption of vegetables high in beta-carotene provides the critical precursor

    necessary for Vitamin A production in the body. Beta Carotene converts to Vitamin A

    as your body needs it. Vitamin A is essential for eyes, skin and proper immune

    function. The levels of carotenoids, such as beta-carotene, in blood and tissue have

    shown consistent inverse associations with various health issues.

    Anthocyanins

    Anthocyanins have antioxidant activity and can scavenge free radicals thereby reducing

    oxidative damage to the cells. The presence of free radicals has been hypothesized bymany as a significant disease causal agent. Anthocyanins from vegetable sources are

    mainly acylated, meaning that they are highly stable due to the inter- and intra-

    molecular copigmentations.

    ORAC Values

    Oxygen Radical Absorbance Capacity (ORAC) values are the most common standard

    of measuring the antioxidant capacity of fruits and vegetables. Foods with a higherORAC value, in general, have a greater antioxidant potential than those with low values.

    There are 6 common harmful reactive oxygen or nitrogen species existing in the body.

    They are:

    1. peroxyl radical (ROO.)

    2. hydroxyl radical (.OH)

    3. hydrogen peroxide (H2O2)

    5. singlet oxygen (1O2)

    6. peroxynitrite (OONO-)

    When these species are exposed to oxidation, free radicals form. Free radicals are

    thought to cause damage to healthy cells. Antioxidants bind with free radicals

    eliminating damaging capabilities. ORAC assay measures the binding capabilities, or

    antioxidant capabilities of natural ingredients compared to a standard, a fluorescent

    probe.

    Cap-e Assay

    The CAP-e assay or Cell-based Antioxidant Protection in Erythrocytes (aka red blood

    cells), is a method for evaluating the antioxidant potential of natural products.

    It is a cell-based assay for the evaluation of whether antioxidants in natural products are

    capable of penetrating and protecting live cells from oxidative damage (free radical

    damage) it is designed to measure the antioxidant bioavailability of a product at the

    cellular level.

    The principle of the method is that antioxidants from the test product enter the red blood

    cells. Unabsorbed antioxidants are removed, and the cells are exposed to a dye. The

    cells are then oxidized and the dye emits fluorescent light in proportion to the amount of

    oxidative damage. A reduction of fluorescence is proportional to antioxidant protection.

    Data from the CAP-e assay can be used to make the following statement: Antioxidants

    in your product can protect live cells.

    ORAC vs. Cap-e

    There are several possible situations pertaining to antioxidant capacity of natural

    products. When comparing data obtained by ORAC versus CAP-e:

    An ORAC value indicates that the tested sample possesses a high potency of

    antioxidant activity chemically. ORAC assays do not determine the

    bioavailability within the body.

    A product may have a high ORAC value but be unable to enter and protect

    cells from oxidative damage

    A product may perform well in both assays

    A product may show better protection of cells than what would be expected

    based on the ORAC value alone
