Waste water analysis with Merck Photometers, test kits and accessories

Analysis of waste water is required by governmental bodies to protect the environment; therefore it is one of the most important topics in industrial and municipal labs.The Spectroquant® photometric system from Merck offers a start to finish solution for safety in waste water analysis.

Essential Equipment

The easy to handle Pharo spectrophotometers combine the advantages of a system photometer on the one hand and the versatility of a spectrophotometer on the other. Reference materials for refractometry

Essential Equipment

Several reference materials for use in testing instruments are described in the European pharmacopoeias (e.g. Ph. Eur. 2.2.6). In

this case, the substances trimethylpentane, toluene and methylnaphthalene are prescribed. This instrument test should be carried out prior to every analysis. Merck now supplies these reference materials. The comprehensive product range includes suitable standards for practically every operating range; the range 1,3915 – 1,6160 is covered.

CertiPUR refractive index standards are suppliedas ready to use kits. They contain five ampoulesof the relevant standard and five ampoules ofwater as reference.

The standards are of course traceable to standard reference materials (SRM) produced by NIST (National Institute of Standards and Technology, USA)) and PTB (Physikalisch Technische Bundesanstalt – Federal Physical-Technical Institute, Germany).

LiChroTest®

Before starting a series of analyses, you need to check if your HPLC system meets with your requirements. These

“Operational Qualification” (OQ) and “Performance Qualification” (PQ) steps involve tests of the different modules on their specifications and a check on the entire system using a real application with regard to the laboratory-specific requirements.

In order to facilitate this instrument qualification in the HPLC laboratory Merck KGaA and VWR International have developed the LiChroTest® products. Thus a time saving operational and performance qualification can be performed routinely and according to standardised methods.

For both, Operational Qualification and Performance Qualification, different test samples for checking precision, accuracy and linearity of the different HPLC modules or the complete system are available. Each set contains several ampoules of sample accompanied by a certificate of analysis which guarantees uniform quality.

For Performance Qualification, the LiChroTest® PQ, a complete test kit for 8 different system tests, is ideal. It comprises test methods, an HPLC column, test samples and a description of the qualification process, including a sample test report. In this way, you can rapidly and routinely carry out fully automatic performance qualification of your HPLC systems. The documentation completed in the course of the qualification procedure is highly useful for passing the next audits.

RNA extraction from cultured cells

Essential Equipment

Expression of genes within cells can be measured by analysing RNA isolated from samples. Here the GeneMole® instrument and

MoleStrips™ RNA Basic Kit are used for automated isolation of RNA from different cells. As no mechanical shearing of tedious pre-treatment steps are needed set-up time for RNA extraction is just 4-5 minutes. MoleStrips™ RNA Basic is developed for downstream reverse transcriptase PCR.

Mole Genetics are now launching pre-filled reagent strip for total RNA extraction from mammalian cells and non-fibrous animal tissue. With the MoleStrips™ Total RNA Basic kit, GeneMole® can extract total RNA from an input of one million cells and up to 10 mg of tissue samples. The RNA yield will vary between cell types. This is due to different size and composition of cells. Both fresh and frozen cell samples can be used. Table 1 shows typical yields for some of the cell types tested.

Table 1: Total RNA yields for a variety of different cell types. The elution volume was specified as 100 µl. UV-spectroscopy was used in quantitate RNA.

RNA extracted with the GeneMole® instrument typically has RIN numbers ranging between 8,5 -10,0. Figure 2 shows an electropherogram and gel-like image of RNA isolated from one million MDCK cells.

Automation is convenient, reduces the risk of injuries caused by repetitive pipetting, limits exposure to chemicals and infectious agents, and gives you more time to focus on other operations.
