• For chemiluminescence, the time to get the image is dramatically reduced and preciousantibody can be saved.

Linearity

The signal response linearity describes therelationship between the detected signal andthe sample quantity within the camera dynamicrange. Linearity is critical for accuratequantification. The greater the linearity, the more preciselyproteins can be quantitated over that range.The Fusion linear range is enhanced by ourunique High Sensitivity Reading (HSR) technologywhich minimizes the noise and increases theimage dynamic. The Fusion is then extremelylinear over its wide dynamic range and caneasily detect large intensity difference betweenbright and faint bands before reachingsaturation.

Spectral Unmixing

Numerous fluorescent dyes are available for multicolor fluorescence imaging. However,interpreting multicolor images can be challenging as the spectral overlap of the excitationand emission characteristics often limits the possibilities to combine these fluorescentmarkers within one sample.

The Fusion Spectral Unmixing is a powerful algorithm designed to correct the spectralcrosstalk between different color channels such as for instance, to separate the Cy2™ signalfrom the Cy3™ crosstalk. With this powerful technique it becomes possible to ascertain thecontribution of different fluorochromes to the total signal and, by chromatic redistribution ofthe intensity, to restore a clear signal for each color channel undisturbed by emission fromthe other fluorochromes. Spectral Unmixing does not create artificially embellished images.The chromatic information inherent to the data is used to redistribute the fluorescenceintensity pixel by pixel while the total pixel intensity is maintained. Thus, quantitative analysisis not only still possible but delivers more significant data.

The SuperResolution with no pixel size compromise

Fusion’s High Sensitivity Reading (HSR) is a unique technologywhich delivers ultra-low noise and sensitivity without alteringthe raw image data. Thanks to a camera modular design, a dualamplifier architecture and a complete control of the electroniccomponents, the camera noise is reduced to a fraction.The HSR technology is a camera “on-head” built-in technology.During the light exposure, the non-necessary cameracomponents are shut down. Once the exposure time is over,the camera components are automatically turned back. Theshut-down process during the image exposition reduced thecamera noise, resulting in a better chip capacity to collect thesignal.

The Fusion FX7 SuperResolution technology providesunparalleled 10 megapixels resolution withoutcompromising read noise, dynamic range, field of view andsensitivity. This means up to 5 times more accurate andmore quantitative data compared to competitors.Our Fusion scientific grade CCD camera has beenspecifically designed for scientific imaging and signalmeasurement. Chemiluminescence and bioluminescence

Fluorescence and multi-fluorescence

Chemiluminescence is the main method used to detectproteins in Western blotting. The chemiluminescentreaction occurs when an enzyme such as horseradishperoxidase (HRP) or alkaline phosphatase reacts with achemiluminescent substrate (such as luminol or dioxetane)to produce a weak signal. The emitted light has awavelength between 400 and 500 nm.The Fusion system is the chemiluminescence imagingmaster thanks to its unrivalled sensitivity and lowest noise.This makes the system ideal not only for Western blot, butalso for luciferase on plants or small animals, and any otherbioluminescence sample.

Fluorescence is the main method used for gene expressionand protein detection. It results from a process that occurs inmolecules known as fluorophores. The fluorophore absorbsthe excitation light, reaching a higher energy state. Byreturning to its former state, it emits fluorescent light. Spectra inside Multimodal imaging platform The Spectra LED light module is designed to add a wide range of applications to your system. With Fusion Spectra, you have 7 excitation wavelengths at your fingertips,ranging from UV to infrared. Up to 4 NET-LEDmodules can be installed simultaneously in thesystem in addition to several UV excitationsources.

The very tight LED spectrum is additionallyconstrained with a very narrow excitation filter.

This means less background in the images ofyour sample and a higher signal to noise ratioto detect the weakest signals.

The FUSION FX Spectra is the first multimodal imaging platform designed for molecular biologylaboratories. It covers chemiluminescence, bioluminescene, fluorescence, multiplexingfluorescence and chemifluorescence applications. Thanks to the Spectra LED modules, thesystem can accommodate up to 7 excitation channels in the IR, NIR, visible RGB and UV area.

The FUSION FX Spectra is ideal for a large array of applications such as Western blotchemiluminescence, Western blot fluorescence, 1D DNA gel, 1D protein colorimetric samples,stain free gel and blot… You can for instance detect two proteins on the same blot usingfluorescence labeling, protein expression can be quantitated relative to a housekeepingprotein without the need for stripping and reprobing. Protein signals can be overlayed so thatboth signals can be visualized simultaneously. Triplexed detection can also be performed forinstance for the detection of two proteins of interest, for example, a protein with and onewithout post translational modification (PTM), normalized to a housekeeping protein on thesame blot.

Each individual light source delivers only a precisely defined range of the spectrum. As aresult, you benefit from extremely high-contrast images – images that will enable you toachieve meaningful scientific results in tasks ranging from simple routine applications tocomplex multiplex applications. The spectral overlap is minimized with the 100nm separationgap of the excitation sources and narrow optical filtering of the emission source. The SL DARQ7 cameraoffers the deepest Peltier cooling availablefrom any Western blot imaging system on themarket, critical for minimization of both noiselevel and sensitivity.In standard, the Fusion SL is equipped with theV.084 lens. The V.084 has unrivalled f/0.84sensitivity and allows the sample to be at 25 cmfrom the camera.

The Fusion SL system is a modular system with extendable capabilities. The built-in capabilityof the system is preliminary chemiluminescence and bioluminescene applications. You canthen upgrade the system if you need UV fluorescence for your gel or Epi-light module for thefluorescence of your blot. The system can grow according to your laboratory need.

The system is also equipped with a 6 positions motorized filter wheel and comes with the AppsStudio and the 3D scan features. Thanks to its modular concept, different options can be addedto the system:

Glossary

Background level: Some degree of noise is always presentin any electronic CCD cameras. Eventhough noise is unavoidable, it canbecome so small relative to the signalthat it appears to be nonexistent. Thedanger is to have the background levelhigher than the signal. In that case, theprotein signal cannot be seen.

CCD technology: CCD cameras are made of sensorstransfer every collected photon into anelectron, namely into digital information.The photons are transferred into pixels.Those pixels will enable thequantification of data.

Excitation filter: Our Spectra LED module (RGB and IR) ismade of a narrow excitation filter. Whenyou excite your sample stained with aspecific fluorophore it is important to usea narrow band pass in order to get asclose as possible to the excitation peakrequired by the fluorophore.

High Sensitivity HSR is a proprietary technology which Reading (HSR) reduces the various source of noise to technology: the lowest floor level so that the lowest

signals can stand out from thesurrounding background.

Multiplexing The multiplexing is the multiple analysis(or multispectral of different type of protein on a single blot fluorescence): or gel. Each protein is stained by one

specific fluorophore that will react withone specific excitation source. The aim isto localize and see the interactionbetween different proteins.

Quadruple Peltier A thermoelectric Peltier transforms an stages: electrical current into a temperature

difference. Four stages enable to achievea fast, deep and uniform cooling.

Spectral overlap The spectral overlap is an undesirable(or cross-talk): effect of excitation of 2 different dyes

sharing the same excitation range. Theoverlap could be compensated using theFusion spectral unmixing algorithm.
