The slicer consists of 3 transparent methacrylate plates (13mm thick, with the middle plate extended to pose as a handle), a thin blade with 102mm cutting edge and blade handle. The base plate is solid and is also the platform in which the fresh tissue is placed. The middle plate has a circular opening in which the tissue platform sits. The cover plate is attached to the middle plate and has a recess of 0.5mm to allow uniformity of the tissue being sliced. The top and middle plates are screwed together when fully assembled. All pieces can come apart for proper cleaning described later in this standard operating procedure.

Reagents The Stadie-Riggs Tissue Slicer as well as individual components can be purchased from Thomas Scientific at http://www.thomassci.com/equipment/microtomes/_/THOMAS-TISSUE-SLICERBLADES. Operational Procedure 1. Place fresh tissue on base plate in marked circular area no greater than 0.6cm in thickness

and 2.8cm in diameter.

2. Secure top and middle plates using setscrews without tightening completely. The “handle” of the middle plate should be held in your non-dominant hand.

3. Carefully pull back the wax paper off of a new blade to reveal the notched end of the blade,

with the beveled end (sharp end) facing away from you. (One optional step is to wash the blade before use to remove any excess oil using a sonicator with mild detergent. Rinse the blade with distilled water and 70% Ethanol. Let dry complety before you use the blade.) Place this exposed, notched end of the blade into the handle and tighten until completely secure.

4. Insert blade with attached handle between top and middle plates, and within the circular outline etched into the two layers of the apparatus. The blade should be facing the tissue as to slice the tissue away from you.

5. Tighten the tissue slicer completely. If the blade is within the circular etching, there should be

enough clearance for it to slide freely. Ensure that the blade is all the way through the middle opening and the handle is up against the tissue holder.

6. Hold the blade handle in your dominant hand and the middle plate in your less dominant

hand. Slice the tissue with a single sweeping motion, where the portion of the blade closest to the handle hits the tissue first and the blade glides through the tissue as you are pulling out and away from you.

7. After you slice through the tissue you can remove the top layer of the tissue slicer apparatus. 8. The slice of tissue on top of the blade, or adhered to the top of the slicer apparatus, is the

piece to be collected (0.5mm thick). This tissue can be used for further processing (biopsy punches, cryopreservation, etc.).

9. Place the blade within the two top plates of the apparatus and retighten the tissue slicer to

repeat.

Cleaning Procedure 1. Dispose of the blade in the appropriate sharps container.

2. Cleaning this instrument from any potentially infectious materials should be done with

caution, within a confined area and according to your University’s Office of Research Safety Guidelines. Rinse the plates and blade handle under water. Use mild detergent and a small brush to get any tissue off of the tissue slicer pieces. Rinse pieces once again.

3. The best way to sterilize the equipment is to place it into a sterile pack and place into an

Ethylene Oxide Sterilizer on the appropriate cycle (the methacrylic plates remain clear). If Ethylene Oxide Sterilization is not available, spray and wipe with 70% Ethanol. Leave to air dry.

• CAUTION: Using harsh chemicals or extended UV time on this equipment may cause the methacrylic plates to become cloudy over time and it will become more difficult to see the tissue while slicing it.

Returning Slicer to Woodruff Lab 1. After 1 week of receiving the equipment, please clean and repackage the Tissue Slicer to be sent back
