It is now over 40 years since lactoferrin was

first isolated. Lactoferrin, which belongs to the

transferring family of proteins, is a 70-80 kDa

single sub-unit glycoprotein and consist of two

domains, each with one iron-binding site that

requires synergistic binding of a bicarbonate

anion. A conformational change in the binding

site, between open or closed forms, accompanies

iron binding and release. The affinity of

lactoferrin for iron is much greater than of

transferrin which functions as an iron carrier in

serum. In the “natural state”, lactoferrin is only

partly saturated with iron (5-30%) and has a

salmon color, the intensity of which depends on

the degree of iron saturation.

It is present in human milk, and many exocrine

fluids (saliva, mucous secretion) as well as

mammalian.

Bovine LF and human LF have approximately

69% of similarities in amino acid residues.

Human milk is the most abundant source of LF. It

is one of the major whey proteins in human milk

with concentrations of 1.4–2.0 mg mL-1 but only

a minor component in bovine milk with only one

tenth (0.1–0.2 mg mL-1) of concentration found

in human milk.

Lactoperoxidase is a hemoprotein that

catalyzes the oxidation of thyocyanate and iodide

ions in the presence of hydrogen peroxide to

produce highly reactive oxidizing agents. These

products have a broad spectrum of antimicrobial

effects. They can kill viruses, gram positive and

gram negative bacteria, fungi, and probably also

mycoplasmas and parasites.

This enzyme is primarily found in such exocrine

secretions as saliva, lachrymal fluid and milk,

cervical mucous and nasal glands. Human milk

does not appear to contain lactoperoxidase

(Tenovuo, 1985). However, a more recent study

confirmed the presence of lactoperoxidase in

human colostrum.

In milk, lactoperoxidase is one of the non

immunoglobulin protective proteins and its

antibacterial activity in raw milk was suggested

as early as 1924.

Bovine milk contains about 20 times higher LP

than that of human milk (Gothefors and

Marklund 1975) and it is the second most

abundant enzyme (De Wit and Van Hooydonk

1996) in bovine milk.

In many ways, lactoferrin has been an easy

protein to investigate; however, there have been

difficulties understanding specific

structure/function relationships, particularly as it

functions in vivo;

Heat stability: At low ionic strength and/or low

pH, the resistance of lactoferrin to heat

denaturation is good and methods for heats

treatment and pasteurization have been

published.

Iron bioavailability: Two features of the

functional properties of lactoferrin are its ability

to bind iron exceptionally tightly and the coupling

of rigid-body domain movements to iron bindings

and release.

Anticarcinogenic effect: Lactoferrin has been

reported to inhibit cancer growth by inducing

apoptosis and stimulating the production of

critical cytokines, such as IL-18 etc.

Antimicrobial agent: Its capacity to bind iron

and to stay bound with iron until pH 3 interfer in

the proliferation of pathogen bacteria.

Antiviral agent: Recently, Antiviral activity has

been described as well for herpes, cytomegalo

and influenzes virus.

Inflammatory and Immune response:

Lactoferrin has various effects on the immune

system, such as regulation of antibody

production, complement activation, and natural

killer cell function.

Anti-oxidant effect: Lactoferrin scavenges

excess iron thereby protects the host to

peroxidation reactions.

� Lactoperoxidase can be combined to

lactoferrin in protection of the mucosal surface

and prevention of tooth decays.

Lactoperoxidase could elicit with bacteriostatic

and/or bactericidal activity on a variety of

susceptible microorganisms including bacteria,

fungi and viruses. The molecular mechanism (s)

of such inhibitory effects depend on the type of

electron donor, test media, temperature and pH

and could range from oxidative killing to

blockage of glycolytic pathways or interference

in cytopathic effects;

Antibacterial Effect: Gram-negative, catalase-

positive organisms, such as pseudomonas,

coliforms, salmonellae and shigellae, are not

only inhibited by the lactoperoxidase system

(LPS) but also, depending on the medium

conditions (pH, temperature, incubation time,

and cell density) may be killed. Gram-positive,

catalase negative bacteria, such as streptococci

and lactobacilli are generally inhibited but not

killed by the LPS.

Antifungal Effect: Seifu et al. declared that LPS

in goat milk inhibit growth and proliferation of

many fungal species viz., Aspergillus flavus,

Penicillium chrysogenum, Trichoderma spp.,

Alternaria spp. and Claviceps spp., Corynespora

cassicola, Phytophthora meadii and Corticium

salmonicolor; however, Candida albicans and

Pythium spp. were not affected by the this

system.

Antiviral Effect: Shin et al. reported the

potential of oral administration of LPS to

attenuate pneumonia in influenza-virus-infected

mice through the suppression of infiltration of

inflammatory cells in the lung.
