IntroductionDefinition Erythrocytes are compatible, biodegradable in nature possessing long circulation half life, and can be loaded with different type of biologically active substance using various chemical and physical methods.

Erythrocytes or RBC’s which have been extensively studies for their carrier ability for the delivery of drugs and drug loaded microspheres at targeted site.

Life span of RBC’s -120 days

Shape- biconcave

Page 4: resealed erythrocytes

Method of isolation of RBC’s

Collect RBC’s of mammalian animals

Add heparin (anticoagulant)

Store at 4°C

Centrifuge

Store at 4-8°C

Use within 2 days for loading

Techniques1. Hypotonic haemolysis

2. Osmotic pulse

3. Electric pulse

4. Chemical perturbation

5. Lipid fusion

Page 7: resealed erythrocytes

Hypotonic haemolysisRBC’s biconcave shape + hypotonic solution of drug

Spherical shape RBC’s

Keep RBC’s in hypertonic solution

RBC’s again take biconcave shape and use as an resealed erythrocytes or loaded erythrocytes

Page 8: resealed erythrocytes

Osmotic pulse

Chemicals are used which improve permeability of RBC’s.

e. g. Urea, AmmoniaRBC’s + isotonic solution of drug

Add chemicals like urea or ammonia

Increase in permeability

Drug diffuse in RBC’s

Store at 4-8°C for 7 days

Page 9: resealed erythrocytes

RBC’s + Isotonic solution of drug Electric current

Increase permeability of RBC’s

Drug diffuse in RBC’s

Store at 4-8°C

Electric Shock haemolysis

Page 10: resealed erythrocytes

Chemical Perturbation

Polyene antibiotics( Amphotericin)

Increase in permeability of RBC’s

Page 11: resealed erythrocytes

Lipid fusionRBC’s + Liposome with drug

Due to concentration gradient drug get diffused in RBC’s
