สถานเสาวภา สภากาชาดไทยไดมการผลตเซรมปองกนโรค พษสนขบาในจ านวนทพอเพยงกบความตองการของประเทศ ตงแตป พ.ศ.2544 และมการสงออกจ าหนายใหกบประเทศเพอนบานบาง ปจจบนสถานเสาวภา สภากาชาดไทยไดน า bulk rabies vaccines (SPEEDA) จากผผลตในประเทศจน (บรษท Liaoning Cheng Da Biotechnology จ ากด, CDBIO) ทผานการรบรองมาตรฐาน GMP มาแบงบรรจทสถานเสาวภา เพอใหมผลตภณฑรกษาและปองกนโรคพษสนขบาครบวงจรส าหรบจดจ าหนายในประเทศ พรอมทงไดมการศกษาทางคลนกทง IM และ ID ในคนไทยเรยบรอยแลว ไดรบอนมตเลขทะเบยนแบงบรรจจากส านกงานคณะกรรมการอาหารและยาตงแตป พ.ศ .2557 และออกจ าหนายภายใตชอวา “TRCS SPEEDA”

การผลตเซรมปองกนโรคพษสนขบา(TRCS ERIG)ในสถานเสาวภา

เรมโดยการฉดวคซนปองกนโรคพษสนขบาในมาทละนอยเพอใหมาสรางภมคมกนโรคพษสนขบาในเลอด (Immunization) จนถงระดบทตองการจงเจาะเลอดมาเพอน าพลาสมามาผลตเซรมปองกนโรคพษสนขบา

สตวทใชส าหรบ Immunization มหลายชนด แตนยมใชมาเพราะสามารถใหพลาสมามาก และสามารถคนเลอดแดงกลบเขาไปในมา ท าใหมาฟนตวเรว (Plasmapheresis) พลาสมาทไดตองเกบในตเยน จะมอาย 2 ถง 3 ป

มาทใชฉด Immunization ตองมสขภาพแขงแรงดงน - อาย 3½ - 10 ป - ปราศจากการตดเชอ ทงเชอจลนทรยและเชอไวรส เชน Equine infectious anemia (EIA) - มการฉดวคซนปองกนโรค - มการตรวจเลอด เพอตรวจสอบทางคลนกเปนระยะๆ - มการถายพยาธเปนระยะๆ - ปราศจากการใชยาปฎชวนะ รวมทงเพนนซลน

Safety and Immunogenicity study of TRCS ERIG

The results showed that TRCS ERIG® is safe with no serious adverse events and no anaphylactic reactions or severe serum sickness were reported.

Determinion of the content of F(ab’)2 and foreign proteins

QSMI has determined the contents of F(ab’)2 and foreign proteins in production batches of TRCS ERIG® produced by current technique and the old methods compared to commercial split IgG chromatographed, commercial whole IgG and crude plasma by using SDS Page and HPLC technique and found that TRCS ERIG® contains high content of F(ab’)2 and high purity (foreign protein content is too small quantity and cannot be detected).

Speeda Production Technology Background

• Virus strain & Cell line

– Vero cells from ATCC; PV2061 fixed rabies virus strain from CDC, USA.

• Cell Culture: Bioreactor Systems

– The main character of the technology is perfusion cultivate the Vero cell on high

density micro-carriers in bioreactor and keep harvesting the virus bulk. It keeps the

environment for cells’ growth to avoid the metabolizing accumulation produced by the

high density cells. The technology of perfusion not only increased the density of cells

grow, but also be helpful to the purification of the product.

• Purification Process: Chromatography

– The continuously harvested virus was concentrated by ultrafiltration and

chromatographic purification was then conducted with chromatography media using

the Sepharose gel system

– It is manufactured under good manufacturing practices (GMP) and strictly fulfills the

WHO recommendations.
