• Unpack the VetScan HM5 analyzer from its shipping container and put out the accesory box.

Note: Store the shipping box, accessory box, and packaging materials in case you need to use them later.

• Place the analyzer on a level surface. • Use the latch to open the side door on the unit, as shown.

• Inspect the unit for any visible signs of damage. • Remove foam 1 (and tape 2 ) as shown below.

Ideal

• Reagent Tubing Kit.

Selecting a Location The analyzer must be installed in a suitable location. • The location should be flat, level, sturdy, vibration-free, and as dust-free as possible.

• Leave at least 50 cm of clearance on all sides of the instrument to allow for adequate airflow and access to connectors.

• Maintain a minimum of 20 cm behind the analyzer’s rear panel for heat dissipation and tube clearance.

• Leave enough space for the reagent pack.

Connecting the Reagent Pack

• Place the reagent pack near the analyzer, either beside or behind the instrument.

• For best results, place the reagent pack at the same level as the analyzer. If you have to place the reagent pack on a lower level, make sure that it is no more than 45 cm below the level of the analyzer’s reagent inlets, as measured from the bottom of the reagent pack to the bottom of the analyzer.

Note: Do not place the reagent pack on a higher level.

Connect the reagent pack as follows. • Open the plastic bag located in the accessory box, and take out the six color-coded reagent tubes and caps with drop-down tubes.

• Attach the supplied connector tubing kit to the instrument, as shown.

• Open the reagent pack box. Open each reagent container (lyse, lyse 2, rinse, cleaner, and diluent) by unscrewing the container caps and piercing the bottle top seals.

• Re-cap each container using the bottle cap with the appropriate color-coded connec- tor and drop-down tube.

• Flip diluent canister 180 degrees prevent reagent errors by ensuring that tubes are not kinked when the box lid is closed.

• Connect each color tube to its matching color outlet. The connectors should snap into each other when properly connected.

• Connect waste bottle.

Initialization of the HM5• Connect Power Supply.• Install paper into the built in printer.• Turn on the analyzer by using the power switch on the upper left of its rear panel.• The analyzer then displays several screens while it runs its software start-up procedure.• Touch Measure, then touch Run.• When prompted, touch OK to run a blank. (Also see “Running a blank”).• The analyzer then initializes its pneumatic system and runs a blank measurement.

• Touch the Measure icon.

• Enter the Patient ID on the “New Sample Screen”.

• Select animal Type.

• Mix the sample well by slowly inverting the tube 10-15 times. Insert the well-mixed EDTA preserved whole blood sample into the appropriate tube adapter, on the sample rotor, and remove the cap.

• Touch the Run button.

If your HM5 is set for autoprint, your results will be printed automatically. If not, press the Print button to print results

Test Results (and blank cycles) are automatically saved to the HM5 database. They can be viewed by pressing the database function key

Analysis is complete in less than four minutes. All results and histograms are automatically saved in the analyzer’s database.

• When analysis is complete, the analyzer displays all measured and calculated parameters, as well as the WBC, EOS, RBC, and PLT histograms:

If the analyzer is not set to print results automatically, press Print to print the results.

Results falling outside of the specified species reference range are marked as follows:

• Results above the reference range are flagged and marked with a plus sign (+).

• Results below the reference range are flagged and marked with a minus sign (–).

• Warning and error flags are marked with upper- and lower-case letters at the bottom.

Running a Blank

A blank must be run every 12 hours. Each blank measurement remains valid for 12 hours of continuous operation, after which the analyzer displays BLANK needed and a new blank is required. In addition, a new blank must be run each time the analyzer has been powered on and off again.

• If the analyzer is in standby mode, touch the measure icon.

• Touch the Run button. • If a blank is necessary, the “Blank needed” screen appears. • Touch OK to confirm and run the blank. • Check the results to see whether the blank falls within the acceptable ranges shown in the following table. If so, press Accept.

Acceptable blanks will have the following readouts:

Running Controls

Controls are run to assess if the analyzer is maintaining calibration.

• Touch the Maintenance icon.

• Touch the Quality control button.

• Touch the QC level (e.g. QC Normal) that will be used.

• Touch References to load the assay and gap values

• Enter QC values (choose one of three methods): a. Enter manually - values can be entered by hand by touching the appropriate fields.

b. Enter by USB pen drive - the QC values file can be loaded by inserting the USB drive into a USB port and touching Load.

c. Enter by Barcode reader - press Load Datamatrix on the Quality Control screen. Scan the 2D barcode on the QC package insert.

• View table of QC databases.

• View QC diagram (for a Levy-Jennings graphic representation of the QC history).

• If the QC results were within range, proceed to running samples. If the QC was out of range, repeat QC run. If the QC is repeatedly out of range, proceed to calibration. (see Calibrating the Analyzer).

Changing the Reagent Pack

• In the Maintenance menu, select Reagent status. Select Replace and then Change pack.

• Select Prime All to prime the reagent tubing.

Calibrating the Analyzer

Calibration is rarely necessary on the HM5, but should be performed when the controls are running out of range.

• Warm up HM5 Normal Control to room temperature. Do not use other level controls for calibration.

• Touch Maintenance and select Calibration. • Touch Measure. Set Sampling Depth to default (--). Press Accept.

• Enter the lot number and assay values from the calibrator package insert and press Accept.

• Mix the Normal Control well by inverting it ten times, uncap the vial and place it into the vial adaptor.

• Touch the Run button. When the results display the screen will show the run as CAL3/1.

• Mix the control well and repeat step 6 twice more.

• After three runs, press the Evaluate button and the screen will display the calibration factors. If there are no errors and the Factors are equal to or between 0.8 and 1.2, press Accept. Consult technical support if there are any errors.

Recommended Maintenance

The VetScan HM5 requires minimal maintenance, but does need attention to proper sample handling and cleanliness to ensure optimal results.

DailyRun a blank.

WeeklyWipe down the HM5 wash head with a damp, lintfree cloth.

With each change of reagent pack Perform Vetscan HM5 bleach cleaning procedure.

Enzymatic CleaningAn enzymatic cleaning is recommended after approximately every 100 tests or at the time of a reagent pack change. Touch Maintenance, Cleaning, then Enzymatic cleaning. Place an EDTA tube with approximately 300 μl of Vetscan enzymatic cleaner concentrate into the sample holder and touch OK.

Shutting Down the Analyzer

Proper shut downs rinse the analyzer to keep it in good condition. So it is important to perform shut downs when the analyzer will not be used for extended periods of time. It is preferable to leave the analyzer on overnight if samples will be run within a couple of days.

Between 3-10 days of non-use

If the analyzer will not be used for more than 3 days but less than 10 days, follow this procedure.

1. Touch the Exit icon.

2. Select shut down, then press accept.3. Power off the analyzer when it emits a tone.

More than 10 daysFollow the instructions below if the analyzer will be shipped or not used for more than 10 days. Detailed instructions will be displayed on the screen.

1. Press the Exit icon.

2. Select prepare for shipment.3. Follow on screen instructions and disconnect the reagent tubing lines and connect the cleaning tube.

4. Put the end of the cleaning tube into distilled water.

5. Power off the analyzer when instructed.
