Diabetes is more commonDiabetes mellitus (DM), or diabetes is a common disease encountered in dogs and cats.

In recent survey, diabetes mellitus occurs in roughly 1 in 200 cats and 1 in 300 dogs.

The diagnosis is often preceded or accompanied by obesity.

What is Diabetes?Diabetes mellitus is an absolute or relative deficiency of the hormone insulin.

Insulin is synthesized and released from beta cells in the pancreatic islets.

Insulin assists with cellular uptake of glucose from the bloodstream into the cells

of the body for energy. In diabetics, glucose isn't transported into the cells and there is

not enough energy for the cells to function normally.

■ Canine diabetes mellitus

• Most diabetic dogs are suspected of having type I DM, characterized by permanent

inability to produce insulin and often required by exogenous insulin administration.

• Most dogs are diagnosed at roughly 7 - 10 years of age.

• Female dogs are at increased risk, regardless of neuter status, than male dogs.

• Neutered male dogs are at greater risk than intact male dogs.

■ Feline diabetes mellitus

• Most diabetic cats have type II DM, characterized by β-cell dysfunction and peripheral

insulin resistance.

• Most cats are diagnosed at > 6 years of age.

• Neutered male cats are at greater risk than female cats.

How much reliable is PT10V Diabetes 4V testing?Fructosamine test performance and reliability

■ Introduction

Fructosamine (FRUC) is a stable complex of blood glucose and proteins produced by an irreversible glycosylation of proteins. FRUC level assesses the average blood glucose concentration over the previous 1-2 weeks, and is extremely helpful in long-term monitoring of diabetic dogs and cats. The PT10V Diabetes 4V provides four different tests of GLU, FRUC, CHOL, and TRIG to manage the increasing diabetic cases in veterinary practices.

■ Conclusion

FRUC test in PT10V Diabetes 4V makes a highly accurate result when FRUC concentration in canine and feline sample is measured.

■ Precision

Material and methods: Precision test was performed by using quality control materials at two different concentrations, Roche Precinorm and Roche Precipath, based on a standard guideline (CLSI EP5-A2). There were 2 replica on a run and 2 runs on a day during 20 days (80 replica in total).

Results: Total precision was calculated by a statistical SW, Analyse-It®. As shown In Table 1, total precisions at both levels are less than 3.0%.

■ Method comparison

Material and methods: Roche P-modular was used as a reference laboratory method, and 4 PT10V devices were used for inclusion of device variation. Canine and feline sera were collected and tested by the reference method and PT10V devices simultaneously. Method comparison was evaluated by a standard guideline (CLSI EP09-A2). Linear regression was done by Passing-Bablok regression, and the bias estimation by Bland-Altman analysis.

Results: The Passing-Bablok regressions of canine and feline samples are shown in Fig. 1 and Fig. 2 respectively. Pearson’s correlation coefficient (R) is 0.9868 for canine samples (N = 104), and 0.9900 for feline samples (N = 78). The mean bias is 0.2% for canine samples, and -0.2% for feline samples.

Therefore, FRUC test of PT10V Diabetes 4V is compa-rable to the reference method, Roche P-modular.

What is PT10V Diabetes 4V testing?When a dog or a cat presents diabetes mellitus with clinical signs of polyuria (PU),

polydipsia (PD), polyphagia (PP) and weight loss, veterinarians prefer to use blood glucose

spot-testing or a glucose curve, fructosamine testing, or a combination of these. In

addition, the veterinarians consider those results along with other valuable risk factors,

such as obesity, dietary causes of hypertriglycedemia.

In this respect, PT10V Diabetes 4V provides four tests of glucose (GLU), fructosamine (FRUC),

total cholesterol (CHOL), and triglyceride (TRIG).

• GLU: A circulating blood glucose concentration.

• FRUC: A stable complex of blood glucose and proteins produced by an irreversible

glycosylation of proteins. A fructosamine level assesses the average blood glucose

concentration over the previous 1-2 weeks.

• CHOL: A part of all lipoproteins as an indicator of abnormal lipid disorder.

• TRIG: A compound of a glycerol and fatty acids as a vital energy source for cell and an

indicator of abnormal lipid metabolism
