Test reagents (TR) and test sera (TS) for the serological identification of Salmonella, Shigella, E. coli& Y. enterocolitica for slide agglutination.
All preparations are medical pro-
requirements of the Directive 98/79/EG (IVD Directive) and therefore carry the CE label. Test reagents which only contain monoclonal antibodies carry SIFIN’s product name Enteroclon.
Enteroclon Anti-Salmonella A-67, omnivalent is used for the initial testing of suspicious colonies to detect the presence of bacteria of the Salmonella genus. As a screening reagent, its function is to record all salmonellaeby rapid and strong agglutination, i.e. a negative result must indicate the absence of Salmonellae to a high degree of probability.
Enteroclon Anti-Salmonella I (A-E) agglutinates only salmonellae of the groups O2 (A) to O 3,19 (E4), and not salmonellae of the groups O 11 (F) to O 67. The Enteroclon is used with isolates that have agglutinated with the omnivalent Enteroclon, to establish membership of the groups A to E. In case of isolates from human test material, this Enteroclon may also be used in place of the omnivalent Enteroclon for initial testing of suspicious colonies for the presence of Salmonella bacteria, as more than 98 % of Salmonella serovars isolated from humans can be assigned to the groups A to E.
Enteroclon Anti-Salmonella II (F-67) agglutinates all salmonellae of the groups O 11 (F) to O 67. This Enteroclon contains no antibodies against salmonellae of the groups O 2 (A) to O 1,3,19 (E4).
Enteroclon Anti-Salmonella Poly-H Phase 1 & 2 is a screening reagent and is used in the serological detection of the H antigens of Salmonella strains. The Enteroclon is a mixture of monoclonal antibodies. It contains antibodies covering the following H antigens or H antigen complexes: a, b, c, d, E, G, i, k, L, r, y, z, Z4, z6, z10, z29,z35, z38, z41 and H 1. Antibodies covering complex antigens, such as Anti-H E, -H L or -H 1, detect all combinations of the respective antigens H e, Hl or H 1. Cross reactions with other genera of the family Enterobacteriaceae have not been found using this Enteroclon.
On their own, positive serological results do not constitute proof that a Salmonella strain is present. For this reason, biochemical tests are required to determine that a sample belongs to the Salmonella genus.
Anti-Salmonella O-Group Pools are intended for use in O-group screening. First carry out the test with Anti-Salmo-nella OMA and Anti-Salmonella OMB: Approx. 98 % of Salmonella can be assigned. If the strain does not agglu-tinate, it is recommended to test this strain with Anti-Salmonella Vi. If this reaction is also negative the strain must then be agglutinated with Anti-Salmonella OMC, Anti-Salmonella OMD, Anti-Salmonella OME, Anti-Sal-monella OMF & Anti-Salmonella OMG.
Anti-Salmonella H-Phase Pools are intended for use in H-phase pool screening. The test, using Anti-Salmonella HMA, Anti-Salmonella HMB or Anti-Salmonella HMC, enables
H-antigens or H-antigen complexes of Salmonella strains. Furthermore the presence of the antigen complex H1 should be tested by Anti-Salmonella H1. If the test reactions are positive, the strain must then be agglutinated with
for further serotyping in accordance with the White-Kauffmann-Le-Minor scheme.
Group-specific Enteroclons facilitate the assignment to the serological groups. The serotype determination is carried

Enteroclons with specificities of low demand and the test sera are lyophilised. The lyophilised products are ready-to-use when dissolved in distilled water (1 ml or 5 ml acc. to the amount given on the label). STEC (VTEC) isolation
Shigatoxin (Stx) producing E. coli (STEC) can cause severe gastrointestinal infections. Fore most among the life threatening complications of an STEC infection are the haemolytic uraemic syndrome (HUS) and thrombotic thrombocytopenic purpura (TPP). While a variety of mobile genetic elements are responsible for the virulence of STEC, their most important virulent characteristic is their ability to produce toxins known as Shigatoxins (Stx) or verotoxins (VT). Stx screening can be done using enzyme immunoassay (EIA) as well as PCR. The Shigatoxin colony immunoblot is a procedure which can be used to isolate Shigatoxin-producing E. coli directly from bacteria

Monoclonal test reagentsare manufactured from culture supernatant of hybridoma and hetero-hybridoma cell lines. Compared with corre-sponding test sera from immune serum donors, these test reagents generally agglutinate more strongly and in many cases also with a higher avidity.
Blood grouping test sera are obtained from immunised donors. All human plasma is tested for the absence of HBs antigen and antibodies against HBs antigen as well as for HIV, and is only used in case the results are negative. Bromelin-RL is also useful in detection of very weak alloagglutinins when determining the ABO blood group.
