Gibbons are smal apes that are threatened to extinction. They are listed in appendix I of CITES. In this study molecular markers were developed to use as identification tool for 3 gibbons Hylobares lor Hylobates pileatus, and Nomascus concolor. Species-specific PCR primers were designec based on polymo~phisms of the mitochondria1 cytochmme b gene. Experiment! demonstrated that optimal magnesium concentration for this primer set was 2.0mM. Increast of annealing temperature in the thennocycling condition clearly increased the specificity 01 all 3 amplification reactions. Eaeh primer pair can specifically amplify its target specie! when amplification was carried out under high smngent condition. No PCR product wen detected when specificity of these primers were tested against human, dog and pig DNA. A simple assay method for characterizing the specificity and efficacy is thus very important especially in drug screening. Estmgen receptor alpha ( E k ) is a superfamily of estrogen receptor, which can interact with ligand and activate cell proliferation and growth of breast cancer. Surface plasmon resonance (SPR) is a powerful technique for detecting an analyte without any labeling. In this study, we have developed the equilibrium condition for observing the specificity of & binding to ERJERE complex on biotin-polyethylene glycol (biotin-PEG) coated surface sensor. To get optimum condition, we used 100 pglml of streptavidin (SA), 0.5 pM of DNA complex and 0.1% ween for preventing non-specific binding. The dissociation rate constant ( k , ) that we investigated was 2 nM. The result was closed to an equilibrium dissociation constant (Kd) 1.8 -+ 0.6 nM of ER interaction with ERE sequences in vitro under equilibrium conditions by fluorescence anisotropy.
