SCIENTIFIC EXPLANATION

The DBC reagent containing the chromogenic substrate

enzyme 5-bromo-4-chloro-3-indolyl-B-D galactopyranoside

(X-GAL)for the detection of ß-

galactosidase (an enzyme indicative of

coliform). In the hydrolysis of ß-Dgalactosidasa.

X-GAL off a chromogenic compound (indigo-blue)

makes the medium turns from yellow to blue-green.

It also contains the fluorogenic substrate of the enzyme4-methyl-umbelliferil-bd-

glucuronide (MUG) for detecting

ß-glucuronidase (an enzyme specific for E. coli). In

hydrolysisß-glucuronidase, MUG clear 4-

methylumbelliferonewhich fluoresces when

exposed to ultraviolet light.The fluorescence distinguishes the presence of E. coli group

coliform.

PROCEDURETake the sample

to the flask to 100ml.

Pour the contents of the powder in the vial with the sample.

Stir until the powder completely integrated.(slightly yellow) Incubate at 35 2 ° C from 18 to 24 hours.

If media is not discolored or yellow there is absence of coliforms.If a change is detected green-blue color indicates the presence of coliform.

CONCLUTION

Our conclution is that with this project we are

showing you how to know if the

water you drink is clean.

Its very important because drinking

contaminated water can make you seek or also

can kill you.Make shure you are

drinking clean water.
