Measure of SRP and TP in extracts

The method is subdivided into two parts;extraction and determination.

2 g of sample extracted with 25 ml 0.1M NaOH (shake over 18 hours); determination of reactive phosphorus (SRP) and total phosphorus (TP) in extracts. elementsDetermination of P-PO4 in conformity with ISO 6878: 2004- Water quality – Determination of phosphorus –Ammonium molybdatespectrometric method.

Principle: Ammonium molybdate and potassium antimonyl tartrate react in acid medium with orthophosphate to form a phosphomolybdic acid that is reduced to intensely colored molybdenum blue by ascorbic acid.

The absorbance is measure at 880 nm between 10-30 min.

• Hydrometer method The sample is treated with sodium hexametaphosphate to complex Ca++, Al3+, Fe3+, and other cations that bind clay and silt particles into aggregates.

• Organic matter is suspended in this solution. The density of the soil suspension is determined with a hydrometer calibrated to read in grams of solids per liter after the sand settles out and again after the silt settles.
