State Lead Advocates
This year’s class of State Lead Advocates will gather for policy and messaging training to support their ongoing advocacy work. State Lead Advocates are committed to building the volunteer advocacy program, motivating existing advocates to engage in the latest action opportunities, and serving as the key contact to the offices of their Members of Congress on public policy issues that directly impact the brain tumor community.
National Events
National event directors and volunteer event committee chairs come together to discuss winning strategies for the national events program. The focus is to foster idea sharing among those who are heavily engaged in community fundraising and awareness across the country. Event committee chairs are committed to representing the National Brain Tumor Society across the nation at our 18 events per year.

NATIONAL BRAIN TUMOR SOCIETY FELDMAN FOUNDER’S AWARD
The National Brain Tumor Society Feldman Founder’s Award is presented to Dr. Darell Bigner, for his invaluable impact on the field of brain tumor research.
Dr. Bigner is the director of the Preston Robert Tisch Brain Tumor Center and the Pediatric Brain Tumor Foundation Institute at Duke University and Leader of the Neuro-Oncology Program of the Duke Comprehensive Cancer Center. He has successfully implemented several internationally recognized programs targeting brain tumors using radiolabeled monoclonal antibodies, immunotoxins, and tumor-specific vaccination. Dr. Bigner and his colleagues have identified several major genetic aberrations in GBM tumors and low grade gliomas, including EGFR, IDH1 and IDH2, and TERT, that provided novel diagnostic and therapeutic implications. They are making huge strides in identifying new drugs and those not previously thought to be active against CNS tumors. Currently Dr. Bigner is overseeing a promising clinical trial utilizing a genetically engineered poliovirus to treat glioblastoma.
The Feldman Founder’s Award recognizes individuals or laboratories for a transcendent scientific discovery, an exceptional body of work, or leadership that had a significant and sustained impact on the brain tumor community consistent with the National Brain Tumor Society’s mission to find better treatments, and ultimately a cure, for people living with a brain tumor. The recipient will receive a grant award and will join the prestigious group of past National Brain Tumor Society Founder’s Award for Research Excellence winners including co-recipients, Dr. Anna Barker, then Deputy Director of the National Cancer Institute and Dr. Kenneth Kinzler, Professor of Oncology at the Johns Hopkins Kimmel Center, for their work on The Cancer Genome Atlas (TCGA); CEO of American Association for Cancer Research, Dr. Marge Foti, PHD, MD (h.c.) for her years of leadership in the field, particularly in systems biology; and Dr. David Louis, Pathologist-in-Chief at Massachusetts General Hospital for his work in the molecular genetic basis of gliomas.

We also continue to foster a robust dialogue with and between community members through social media, online community chats, and events across the country. During Brain Tumor Awareness Month, we heard from survivors, caregivers, family, and friends who shared advice and knowledge about brain tumors to educate and empower others. On Facebook, people share their experiences, questions, and stories while continuing to stay attuned to what is going on in the community.
The important thing to always remember is that together we are stronger than this disease!

NATIONAL BRAIN TUMOR SOCIETY PROGRAMMATIC OVERVIEW
Our strategic initiatives and funded programs aim to improve the understanding of brain tumors to transform research findings into new and more effective treatments. Using an integrated approach, we strive to address obstacles from the very first phases of basic research to the clinical trial and FDA approval process. Some of these efforts require funded research programs, some require new public policy solutions, and many need to combine tactics. All require getting the right parties together; including researchers and clinicians, biopharmaceutical and other for-profit industry companies, government funders, Congress, regulators, other philanthropic and nonprofit organizations, or some combination thereof.
The following pages provide information on each of our initiatives and currently funded researchers. In addition, we continue to foster collaboration by hosting workshops and meetings covering critical brain tumor topics, participating in key discussions taking place in the community, and mobilizing volunteer advocates across the country.

CLINICAL TRIAL ENDPOINTS INITIATIVE
This Initiative leverages both our research and public policy programs to work with the US Food and Drug Administration (FDA), patient advocacy and philanthropic organizations, medical imaging, biotechnology, and pharmaceutical companies, neuro-radiologists, neuro-oncologists, contract research organizations, and the National Cancer Institute to create change in the brain tumor clinical trial process.
As part of this effort, the Jumpstarting Brain Tumor Drug Development Coalition (National Brain Tumor Society, Accelerate Brain Cancer Cure, Musella Foundation, and Society for Neuro-Oncology) was created to convene these key stakeholders in two separate workshops to advance alternative endpoints for brain tumor clinical trials. The workshops represent the state of brain tumor endpoint measurement and contribute to the development of new criteria required for designing valid clinical trials using alternative endpoints. This work promises to create more incentives for future research and investment in brain tumor treatments and deliver a pipeline of new therapies to the community.
DEFEAT GBM RESEARCH COLLABORATIVE
The Defeat GBM Research Collaborative, a subsidiary of the National Brain Tumor Society, is a bold, multi-faceted, and concentrated effort, which aims to double the five-year survival rate of GBM patients – in just five years.
The following synergistic and simultaneous research projects are underway:
Aim 1: Finding the Right Target
• Ludwig Cancer Research, Dr. Web Cavenee and Dr. Frank Furnari Along with identifying the right biological target(s), this project seeks to understand any associated resistance mechanisms.
Aim 2: Finding the Right Drug
• The University of Texas MD Anderson Cancer Center, Dr. Fred Lang and Dr. John de Groot Accelerate drug development by identifying existing molecules/drugs that can be applied specifically to GBM, both on their own and in combination, as well as develop new therapies.
• Memorial Sloan Kettering Cancer Center, Dr. Ingo Mellinghoff Better understand a tumor’s systematic molecular and biological response when attacked with clinical inhibitors of well-characterized GBM mutations. Use systems biology approaches to develop and test rational combinations of inhibitors against these known GBM drug targets.
Aim 3: Determining the Right Patient
• Ludwig Cancer Research, Dr. Paul Mischel and Dr. Tim Cloughesy Develop predictive biomarker panels to guide therapy – including combination therapy – for each patient based on the molecular composition of their tumor.

2014 NATIONAL BRAIN TUMOR SOCIETY GRANT RECIPIENTS
PEDIATRIC RESEARCH
National Brain Tumor Society is committed to directing one-third of its research budget to pediatric-specific initiatives. Most recently we funded work in molecular profiling, which made important contributions to the field. Currently work in Developmental Neurobiology is underway. Looking ahead, Project Impact is the National Brain Tumor Society’s new, multi-faceted initiative to address the key interrelated barriers that are slowing drug development for pediatric brain tumors, specifically high-grade gliomas. Project Impact will combine efforts in funded research and public policy to confront inefficiencies that currently exist in both the pediatric brain tumor pre-clinical research system and pediatric cancer clinical trial environment.
Robert Wechsler-Reya, PhD Sanford-Burnham Medical Research Institute Genetic targeting of cerebellar stem cells to study development and tumorigenesis
The most aggressive subtype of medulloblastoma occurs in tumors that have increased expression of the MYC oncogene. In this research, the Wechsler-Reya laboratory will build on previous research that suggests that stem cells can give rise to MYC-driven medulloblastoma. The researchers will create and study mouse models that allow targeting of genes to stem cells and then follow the course of normal stem cells and generate models of MYC-driven medulloblastoma. The goal is to discover potential new treatments based on an understanding of the tumor cells of origin.
Alexandra Joyner, PhD Memorial Sloan-Kettering Cancer Center Identification of genes and cell behaviors regulated by Shh/Gli2 signaling in the cell of origin of a major subtype of medulloblastoma using novel genetic tools in mouse
Medulloblastoma, the most common brain tumor in children, occurs in multiple molecular subtypes. A process called hedgehog signaling (named after the hedgehog gene Hh) is elevated in tumor subtypes that arise from cerebellar granule cell progenitors (GCPs). In this research, the Joyner laboratory will investigate how altering the level of signaling in GCPs in genetically engineered mice alter GCP behaviors. The goal is to discover novel signaling pathways that are important for normal cerebellum development.

PEDIATRIC RESEARCH (CON’T)
Joseph Scafidi, DO Children’s Research Institute The effects of molecularly targeted therapies on the developing neurogenic niches
Although recent treatment compounds have shown promise in decreasing the burden of pediatric brain tumors, the compounds also affect the developmental pathways of a normal developing brain, and long-term effects are unknown. In this study, the Scafidi laboratory will assess the cellular and functional effects of compounds in rodents at various developmental ages, and whether and rehabilitation efforts after treatment can induce brain plasticity and address the deficits associated with treatment. Some of his early findings were published in the prominent journal Nature.
INNOVATION GRANT - PEDIATRIC
David H. Gutmann, MD, PhD Washington University School of Medicine KIAA1549:BRAF genetically-engineered mice for pediatric glioma-targeted therapeutic drug discovery and evaluation
One of the most common brain tumors in children is the pilocytic astrocytoma. This tumor is often caused by either genomic rearrangement of the BRAF gene (sporadic) or the inactivation of the NF1 tumor suppressor gene (inherited) that, when mutated, causes neurofibromatosis type 1. For this research project, the Gutmann laboratory will conduct experiments using advanced systems biology bioinformatics to identify potential biological treatment targets for both sporadic and inherited pilocytic astrocytoma. He also created a new model with the BRAF mutation that did not previously exist.

OLIGODENDROGLIOMA RESEARCH FUND
The Oligodendroglioma Research Fund is part of our Community Research Fund program – an initiative allowing contributors to pool donations to support research on a specific type of tumor. Donations large and small can be earmarked and combined to fund a two-year grant of $300,000, the minimum grant amount necessary to conduct significant research studies.
David Louis, MDMassachusetts General Hospital (MGH) Mapping the Epigenomic Landscape of Oligodendroglioma to Elucidate Tumor Growth and Chemosensitivity
Dr. Louis and his team will seek to identify the elements that drive the growth of oligodendrogliomas by applying next-generation sequencing tools and experiments. Dr. Louis’ work is designed to lead to discoveries of specific molecular targets, which subsequent studies would examine for potential therapeutic approaches.
Anders Persson, PhDUniversity of California, San Francisco IDH1R132H-induced Transformation of Oligodendrocyte Progenitor Cells into Oligodendroglioma
Dr. Persson and his team will seek to create predictive disease models (in mice/human/xenograft tissue) to prove that a specific gene mutation of the common enzyme IDH1 creates a molecule, called oncometabolite 2-HG, that facilitates the transformation of normal oligodendrocyte cells (OPC), a common cell in the human brain, into malignant, oligodendroglioma cells. Dr. Persson’s team will work to validate that these OPC’s are the “cell of origin”, and know what mutations drive them to develop into an oligo tumor, in order to develop targeted therapies to attack their pathways.
Robert Jenkins, MD, PhD & Dr. Daniel Lachance, MDMayo Clinic Function Biology of Germline Predisposition to Oligodendroglioma
Drs. Jenkins and Lachance will seek to generate both cell lines and animal models that replicate oligodendroglioma from initiation, based on previous discoveries of germline variations that contribute to tumor development. Current models reflect oligodendroglioma late in their course, and models from disease initiation may allow therapeutic screening for treatments effective against the disease in its earliest and most treatable state.
