TECHNOLOGY DEVELOPED

DEVELOPMENT OF A THERMOSTABLE ORAL PELLET VACCINE AGAINST NEWCASTLE DISEASE

Newcastle Disease (ND) is a highly contagiousviral disease affecting poultry of all ages. In India thedisease is present in an enzootic form and frequentoutbreaks cause devastating losses not only tocommercial poultry but also to small scale farmerswho depend on backyard poultry for their nutritionalneeds and much needed cash income in times of need. The disease can be controlled by the use of vaccinesand most of the available vaccines are unsuitable foruse in villages as they require maintenance of coldchain for transportation to long distances and areavailable in large doses. A project initially funded bythe Indian Council of Agriculture and now funded bythe Global Agency for Livestock and VeterinaryMedicines (GALVMed) of the United Kingdom for

further research wasinitiated at theDepartment of VeterinaryMicrobiology, MVC todevelop a thermostableoral pellet vaccine againstND. A total of 160 local samples from village chickens which were never vaccinated

were collected in the year 2000 and 16 ND virus isolates were obtained. All the isolates were screened forvirulence by Intra-cerebral pathogenic index (ICPI) and one of the isolates, D58 of low virulence with an ICPIvalue of 0.14 has been used for further study and vaccine production. The isolate was thermostabilised byexposure to 250C for 36 days, 370C for 29 days and 560C for 30 minutes. Based on amino acid sequence of thefusion protein cleavage site (FPCS), HN and F gene sequence analysis and phylogenetic studies, the isolate wasfound to be of low pathogenic type and was positioned in groups with other low pathogenic and apathogenicviruses. The master (MSV) and working seed viruses (WSV) were prepared and standardized as per thespecifications of the OIE (2000). From the WSV, an oral pellet vaccine was prepared. Extensive laboratory andfield trials were conducted with the oral pellet vaccine. The vaccine is regularly supplied to the KVKs for onfarm trials. A little more than 75,000 doses have been tested at the field. The oral pellet vaccine was found tobe safe and potent as per the norms prescribed by FAO (1991) and OIE (2000). The vaccine producedprotective serum antibody titre of 26 at laboratory and field trials which is above the value of 23 suggested byFAO and OIE. The vaccine also produced satisfactory immunity for a period of three months. No post-vaccination complications like respiratory distress or dullness or off-feed and no occurrence of ND in any ofthe vaccinated flock despite the fact that still ND is prevalent in this part of the country have been reported. The vaccine has been found to produce humoral, cell mediated and mucosal immune responses in laboratorytrials and found to give protective immune response in virulent virus challenge. Dr. J. John Kirubaharan,Professor the Principal Investigator of the scheme developed the vaccine.

NEW SCHEMES

The Government of Tamil Nadu accordedsanction for the implementation of the componenton “Fodder seed production” under the StateFodder Development Scheme at two locations ofTANUVAS viz. IAN, Kattupakkam and TANUVAS

SRRC, Tirunelveli for a period of one year(2011-2012) at a total cost of Rs.123.20 lakhs(Rs. 61.60 each). The project will be implementedthrough the Director of Animal Husbandry andVeterinary Services.

Story of a beneficiary..........

Tmt. K. Kala of Palavankudivillage, Sivagangai districtstarted a small poultry farm infree range system in her fiveacre agriculture land. As herland falls under rain fedagricultural belt of Tamil Naduonly one cropping is possible. The small poultry farmprovides the extra income for her daily familylivelihood. But outbreak of diseases especially Newcastledisease have caused heavy losses to her income. Thoughshe is aware of vaccination, she is unable to protecther flock as she has to take them to a veterinaryhospital or get the help of a vaccinator. When shecame to know about the oral pellet Newcastle diseasevaccine through KVK, Kundrakudi, she adopted thenew technology and gave the vaccine to her flock. Now there is no mortality in her farm.
