    Mast cells participate in some types of inflammatory reactions involvingchanges in the microcirculation of certain tissues. Among the known vasoactivesubstances of importance, histamine has been found in mast cells (1) of severalspecies of animals and in certain species serotonin is also present (2). A varietyof substances (the formaldehyde polymer of p-methoxyphenethylmethylamine(48/80), ovomucoid, and dextran) when administered to the rat elicit aninflammatory response, cause histamine and serotonin release and the morpho-logical change of degranulation (3, 4). No clear description of the processinvolved in release of the active amines and other mast cell constituents hasyet been presented. A major obstacle has been the lack of an appropriate wayof observing the action of various agents on the structure of the mast cell andits constituents under controlled conditions. Hence, a new method of observa-tion was sought to study the mechanism of mast cell secretion in vitro.

    Certain properties of Millipore filters made these suitable for use in thesystem: biological inertness, relatively high filtration rates, appropriate porediameters for retaining mast cells, and transparency following dehydrationand clearing. This paper presents a method developed for in vitro study of mastcells. The method permits observations of cell morphology and measurementsof such substances as histamine and serotonin under a variety of experimentalconditions. Application of this method has revealed significant differences inbehavior among agents known to liberate histamine and cause degranulationof mast cells in vivo. Thus, 48/80 and polymyxin B act directly on mast cells,while ovomucoid and dextran have no discernible effect on isolated mast cells.
