LaPrep Sigma – the new compact preparative chromatography system from vWr
Being a truly modular system with many possible configurations, having the smallest
foot print in the market, an easy to use touch screen panel and ethernet connection, it is
the ideal preparative hPLC system for modern purification.
Running on the intuitive eZChrom platform, the new laprep sigma has capabilities ranging from analytical to preparative scale, pump capacity from 1 to 600 ml/min as well as easy connection to a fully automated fraction collector. Along with the option of a single or multiple wavelength uV detector and options for autosampler or single injection sample delivery, the new laprep sigma from VWR is an excellent choice for any prep lab whatever the configuration requirements.

Pumpseach pump head is manufactured using titanium for all sample contact components. this makes laprep sigma pumps suitable for use with biological samples where a high salt concentration buffer would otherwise corrode stainless steel. All pumps are double piston with electronic residual pulsation suppressor and active piston seal wash which are fitted at the site of manufacture and tested before shipping.
the capacity range of the pump heads, with maximum flow rate for each at 50, 200 and 600 ml/min, means that it is highly flexible for use in all chemistry laboratories from bench use in research laboratories to scale up laboratories. As shown in table 1, with its range of flow rates, the laprep sigma covers loading capacity of up to 5 g and loading volume of 80 ml. this flexible flow rate capacity coupled with the modular design means it can be a really cost effective solution.

Configuration and softwareRunning on the intuitive eZChrom platform, the new laprep sigma is a compact preparative chromatography system from VWR. being a truly modular system, with the smallest footprint in the market, an easy to use touch screen panel and ethernet connection lapre sigma is an ideal addition to any laboratory needing a flexible prep solution.
the laprep sigma has capabilities ranging from analytical to preparative scale, pump capacity from 1 to 600 ml/min, as well as easy connection to a fully automated fraction collector. Along with the option of a single or multiple wavelength uV detector and options for autosampler or single injection sample delivery, the new laprep sigma from VWR is an excellent choice for any prep lab whatever the configuration requirements.
As with all VWR’s chromatography instrumentation, the new laprep sigma is designed with robustness and durability in mind.
fraction and system control with eZChrom elite™
the laprep sigma can run in a semi-prep mode with the 50 ml/min pump. in addition, because it is controlled by eZ Chrom software it can be combined with the VWR hitachi Chromaster analytical system in order to provide a simple, easy to use semi-prep system whilst taking advantage of the Chromaster’s excellent autosampler and detector capabilities*.
the eZ Chrom software allows for straight forward peak recognition. it starts to search for substance peaks at a pre-determined time. the substance peaks are recognised by looking for criteria which fulfil the peak start and end level. this leads to a simplification of the programming needed for a complete fraction as only one line needs to be programmed for a complete fraction with the same parameters.
the peak recognition and subsequent fraction collection can clearly be seen in figure 1. the eZChrom elite™ software using the eZ prep Driver clearly displays colour coded areas of the chromatogram to represent peak collection, peak recycling or solvent recycling. figure 1 shows the “signal level / slope” command in operation for fraction collection.

Direct control of fraction collectionit is also very simple to directly control either the fractionation valve or fraction collection via the direct control tool in eZ prep. figure 2 shows the control panel for the fractionation valve. With only a few clicks of the mouse, the flow can be diverted into the corresponding fractionation vessel until a further command diverts the flow to another collection vessel or to waste.

When trying to separate two or more components that elute at similar times with an overloaded column, peak recycling can be set-up, where by unresolved fractions are fed back into the system in order to undergo further separation. this is particularly suited to processes where only one fraction is of interest and a more environmentally sound separations process is being sought.
With preparative chromatographers in today’s laboratories being required to provide ever more bespoke and tailored solutions to medicinal chemists, materials chemists and chemical engineers, VWR has designed a highly flexible system to suit the vast majority of cases.
