PERHITUNGAN DOSIS
Perhitungan dosis Andriol (Testosteron Undekanoat)
Dosis andriol (Testosteron Undekanoat) untuk pria 70 kg adalah 80 mg
(Adhi D., Aulia S., Azrul A., Handaya, Merdias A., Rianto S., dkk., 2005)
Kandungan andriol (1 tablet) = 40 mg
Faktor konversi untuk mencit yang beratnya ± 20 g = 0.0026 (Paget & Barnes, 1964)
Mencit yang digunakan pada penelitian beratnya ± 33.4 g
Volume lambung mencit ± 0.5 ml
Perhitungan :
Dosis Testosteron undekanoat yang dikonversikan untuk mencit
= 80 mg x 0.0026
= 0.208 mg / mencit 20 g
Dosis Testosteron undekanoat yg diberikan untuk mencit
= (33.4 : 20) x 0.208
= 0.3474 mg/mencit 33.4 g
= 10.4 mg/kgBB mencit/0.5 ml
Pembuatan larutan Testosteron undekanoat = 0.3474 mg/0.5 ml
= 0.6948 mg/ml
= 6.948 mg/10 ml
= 6.9 mg/10ml
40 mg/tablet = 40 mg/10ml
= (6.9 : 40) x 10 ml = 1.7 ml
Larutan Testosteron undekanoat akhir yang diberikan:
1.7 ml Testosteron undekanoat dalam Na-CMC 1% + 8.3 ml Na-CMC 1%.

Perhitungan dosis infusa rimpang lengkuas merah
Dosis pada manusia adalah 50 g (Admin, 2005)
Faktor konversi untuk mencit yang beratnya ± 20 g adalah 0.0026
Mencit yang digunakan pada penelitian beratnya ± 33,4 g
Volume lambung mencit ± 0.5 ml
Perhitungan:
50 g rimpang x 0.0026 = 0.130 g / 20 g mencit
= 0.217 g / 33,4 g mencit
= 217 mg / 33.4 g, setara dengan 1 x DM (dosis manusia)
IRLM-1 :
Dosis infusa yang digunakan 43,4 % = 43.4 g / 100 ml
= 434 mg / ml
434 mg / ml sama dengan 217 mg / 0.5 ml (volume lambung mencit)
217 mg rimpang dilarutkan dalam 0.5 ml aquadest.
Jadi dosis yang digunakan 217 mg rimpang setara 1 x DM (dosis manusia)
IRLM-2:
Dosis infusa yang digunakan 65.1 % = 65.1 g / 100 ml
= 651 mg / ml
651 mg / ml sama dengan 325.5 mg / 0.5 ml (volume lambung mencit)
325.5 mg rimpang dilarutkan dalam 0.5 ml aquadest.
Jadi dosis yang digunakan 325.5 mg rimpang setara 1.5 x DM (dosis manusia)

Pembuatan infusa
Untuk infusa rimpang 65.1% dibuat dengan cara 65.1 g rimpang yang telah
dikeringkan, diblender sampai halus kemudian dilarutkan dengan aquadest sampai
100 ml dalam panci infusa kecil dan dipanaskan selama 15 menit sampai 900 C
(dimasukkan ke dalam panci infusa besar yang diisi air dan sudah dididihkan).
Diberikan infusa per oral 0.5 ml / ekor yang mengandung 325.5 mg rimpang.
Untuk infusa rimpang 43.4% dibuat dengan mengencerkan infusa rimpang 65.1%,
dengan cara 10 ml infusa rimpang 65.1% ditambahkan 10 ml aquadest hangat
sehingga konsentrasinya menjadi 43.4%. diberikan infusa peroral 0.5 ml / ekor yang
mengandung 217 mg rimpang.

PROSEDUR KERJA
H 1 : Pembelian 84 ekor mencit (24 ekor jantan, 60 ekor betina) dan tanaman
lengkuas merah (rimpang).
H 2 – 6 : Aklimatisasi mencit dan pengeringan tanaman serta pembuatan serbuk
rimpang.
H 7 – 13 : Persiapan larutan Na-CMC 1% 0.5 ml per mencit (6 ekor mencit) →
kontrol.
Pembuatan larutan Testosteron Undekanoat (TU) dosis 10.4 mg/kgBB
dalam 0.5 ml larutan Na-CMC 1% per mencit (6 ekor mencit) →
pembanding.
Pembuatan IRLM-1 dan IRLM-2 → bahan uji (perlakuan).
T 0 : Pemberian perlakuan 0.5 ml peroral dan penempatan di sudut kotak
yang diberi sekat (H 11 & 13)
Percobaan :
Hewan uji yang terdiri dari 24 ekor mencit jantan yang dibagi dalam 4 kelompok
Kelompok I : terdiri dari 6 ekor mencit menjadi perlakuan IRLM-1
Kelompok II : terdiri dari 6 ekor mencit menjadi perlakuan IRLM-2
Kelompok III : terdiri dari 6 ekor mencit menjadi perlakuan kontrol
Kelompok IV : terdiri dari 6 ekor mencit menjadi perlakuan pembanding.
T 45 : mencit betina di tempatkan di sudut kotak lainnya
T 60 : sekat dibuka
