Reversed PhaseA. C18 is the most popular bonded phase due to i ts versat i l i ty anddurabil i ty. Most separations may be accomplished to at least someextent with C18. The separation is achieved by exploit ing differ-ences of hydrophobicity within the sample matrix. C18 wil l alsotend to exhibit greater loading capabil i t ies. This is usually the bestchoice for a non-polar to medium polar sample matrix with l i t t le orno other avai lab le in format ion.

B. The C8 phase conta ins a shor ter a lky l chain than C18, resul t ingin a sl ightly different selectivity. The separation is st i l l obtainedthrough hydrophobicity differences within the sample matrix, but i toccurs to a somewhat lesser degree than C18, result ing in a reducedretent ion. Wi th a sample exhib i t ing too much retent ion on a C18column, the C8 is an excellent alternative. The lower capacity of theC8 column means shorter analysis t imes and greater lab eff iciency.

Normal PhaseC. The si l ica phase contains no addit ional bonded chemistr ies toaffect the resultant chromatography. The separation mechanismsat work on a normal phase s i l ica co lumn inc lude type, or ientat ion,and quant i ty o f po lar funct ional groups wi th in the sample matr ix .Sil ica columns are best uti l ized with polar samples. The retentionsimilari ty between TLC plates and si l ica phase columns tends tosimplify method development for sample analysis.
