Image particle analysis: 

Open image or drag and drop. There are two way to go for the particle analysis. ImageJ want either the image is binary which can be done by

For our purpose the other way is the best. The threshold and then the particle analysis As shown below.

Choose the required level for threshold e.g. the white area. 

 

Then go to particle analysis

Repeat same thing for the black area by choosing the appropriate threshold.

In the  summary the total area in calculated for white or black areas. At the end one has the white and black area counted and listed. By subtracting these two added sum from the total picture area one gets the gray area. This procedure should be repeated 4‐5times to get the average value and the standard deviation.
