Since the majority of disease genes in humans are present in flies, studying flies and the mechanisms that operate in these organisms becomes important and useful. Indeed, in the past we have been able to use fly genes to make human cells more resistant to certain stresses.

Project 1 – P.I. Core A – P.I. Core C – Co-Inv This PPG is dedicated to the understanding of how tissues die or survive when animal or human tissues are deprived of oxygen. This is basic for many disease states such as obstructive sleep apnea, and cardio-respiratory diseases (e.g., Asthma and Sickle cell disease as well as COPD). We employ two animal models, one being susceptible and the other tolerant to low O2 to understand the differences and be able to render mammalian tissues that are sensitive to low O2 much more resistant to it. 1R01GM089897 (Haddad P.I.) 7/1/10-6/30/15 NIH Total Support: $1,250,000 Genetic Dissection of Hypoxia Tolerance This application addresses questions that will lead to our basic understanding of how flies, which are tolerant to low O2, manage to escape from cell and tissue damage when they are exposed to low O2. Furthermore, when we develop a framework for our understanding of tolerance and learning from flies, we will engineer cells or tissues from mammals that will be much more tolerant to low O2.

In order to be consistent, and because cortical neurons have been shown to be vulnerable to hypoxic exposure, this work will be focused on the neocortex.
