UJI AKTIVITAS EKSTRAK ETANOL BAYAM DURI (Amaranthus spinosus L.)

TERHADAP BAKTERI Staphylococcus aureus DAN Pseudomonas aeruginosa

DENGAN METODE DIFUSI AGAR

ABSTRAK

Tumbuhan Bayam duri (Amarhantus spinosus L.) secara empiris digunakan untuk mengobati

berbagai penyakit diantaranya eksim, disentri dan diare. Aktivitas antibakteri dari daun bayam

duri ini masih belum banyak diteliti. Tujuan penelitian ini dilakukan untuk mengetahui

aktivitas antibakteri serta menentukan nilai Konsentrasi Hambat Tumbuh Minimum (KHTM)

dari ekstrak etanol bayam duri (Amarhantus spinosus L.) terhadap bakteri Staphylococcus

aureus dan Pseudomonas aeruginosa. Penelitian ini meliputi ekstraksi daun bayam duri,

penapisan fitokimia, kromatografi lapis tipis terhadap ekstrak bayam duri, uji aktivitas

antibakteri, penentuan Konsentrasi Hambat Tumbuh Minimum (KHTM) dengan metode

difusi agar dan uji banding dengan ekstrak daun sukun (Artocarpus communis Forst.). Hasil

penelitian menunjukkan bahwa ekstrak etanol daun Bayam Duri (Amaranthus spinosus)

memiliki aktivitas antibakteri terhadap Staphylococcus aureus , sedangkan pada

Pseudomonas aeruginosa tidak memiliki aktivitas. Kosentrasi Hambat Tumbuh Minimum

(KHTM) ekstrak etanol daun Bayam Duri (Amaranthus spinosus L.)terhadap Staphylococcus

aureus berada pada rentang 6000 – 7000 ppm. Hasil uji banding daun bayam duri

(Amarhantus spinosus L.) dengan ekstrak daun sukun (Artocarpus communis Forst.) terhadap

Staphylococcus aureus sebesar 1 : 54,075

ABSTRACT

Spinach spine plant (Amarhantus spinosus L.) had been empirically used to treat various

diseases including eczema, dysentria and diarrhea. Antibacterial activity of Spinach spine

plant is still not much studied. The purpose of this research is conducted to determine the

antibacterial activity and determine the value of the Minimum Inhibitory Concentration (MIC)

of ethanol extract of spinach spine plant (Amarhantus spinosus L.) against Staphylococcus

aureus and Pseudomonas aeruginosa. This research involves the extraction of spinach spine

plant, phytochemical screening test, thin layer chromatography of spinach spine plant extract ,

antibacterial test against Staphylococcus aureus and Pseudomonas aeruginosa, determination

of Minimum Inhibitory Concentration (MIC) using diffusion method, and comparison with

extract of breadfruit leaves (Artrocarpus communis Forst.). The results showed that spinach

spine plant extract had antibacterial activity against Staphylococcus aureus but did not has

antibacterial activity against Pseudomonas aeruginosa. The Minimum Inhibitory

Concentration of ethanol extract of spinach spine plant(Amaranthus spinosus L..) extract

against Staphylococcus aureus was on concentration range 6000 – 7000 ppm. The ratio of

antibacterial activity between spinach spine plant(Amaranthus spinosus L.) extract and

breadfruit leaves (Artrocarpus communis Forst.) extract against Staphylococcus aureus was

at 1 : 54.075.
