生物 A (Organism A) は、細胞質の中に数十個の核 (nucleus) を持つ単細胞生物です。生
物 A は、土の中でバクテリアを食べて成長し、2 mm ほどの大きさになるとすべての核が
同時に分裂し、引き続いて細胞質が分裂します。研究室内でも、バクテリアを与えることに
より寒天培地上で増殖させることができます。
S さんは、遺伝研構内の土から単離した生物 A を培養しました。生物 A には黄色のもの(Y
型)と緑色のもの(G 型)がいますが、Y 型の 1 個体を増殖させたところ、子孫のほとんど
は Y 型でしたが、少数の G 型の個体も混じっていました。一方、G 型の個体1個を単離し
て増殖させたところ、子孫の大部分は G 型でしたが Y 型の個体も少数混じっていました。
これらの集団をさらに注意深く観察したところ、細胞分裂直後にのみ、ごく低い頻度で Y
型の個体が G 型に転換したり、G 型の個体が Y 型に転換したりするのが観察されました。

問1 S さんは Y 型から G 型、G 型から Y 型の転換頻度を知りたいと思いました。あなた
ならどのような方法で転換頻度を計りますか?実験をデザインしてください。
問2−1 Y 型から G 型への転換頻度は 1 回の分裂あたり1%、G 型から Y 型への転換頻度
は 1 回の分裂あたり 2%でした(どちらも15℃の培養での計測。)。培養を長く続けている
と集団の中の Y 型と G 型の比率は一定になりました。G型と Y型が同じ速度で分裂する(一
定時間内に同じ回数分裂する)と仮定して、定常状態での Y型と G型の細胞数の比を予測し
てください。また、計算の過程を筋道立てて説明してください。
問2−2 問2-1で述べた集団を30℃に移して長く培養したところ、ほとんど全てが G
型の個体になりました。この現象の説明の一つとして、15℃から30℃への温度変化によ
り転換効率が変化した(すなわち、Y 型から G 型への転換頻度の増加や G 型から Y 型への
転換頻度の減少が起きた)可能性が考えられます。現象を説明できる他の可能性を提案して
ください。できるだけ複数の可能性を挙げてください。

問3−1 S さんは、生物 A が Y 型から G 型、G 型から Y 型に可逆的に転換できるメカニ
ズムに興味を持ちました。Y型と G型の間の転換はどのようなメカニズムで起こるか、考え
られる仮説を一つあげてください。

問3−2 問3-1で挙げた仮説を検証するための実験や解析を計画してください。
問4 生物 A を用いて、あなたなら他に何を調べたいですか?できるだけ異なった二種類

Read the following story and answer all the questions. This examination is aimed at measuring
your abilities in logical thinking and writing (including drawing schematics). The setting of
these questions is basically imaginary. You are free to choose experimental settings unless they
are given. Please note that you are required to define the settings you chose clearly in your
answers.

Therefore, even if you cannot answer a question, you may be able to answer subsequent questions. * Please write and draw your answers within the assigned spaces in the answer sheet.
Organism A is unicellular, but each cell body contains tens of nuclei. Organism A grows in the
soil by eating bacteria. When it reaches about 2 mm in length, all nuclei divide simultaneously
and subsequently the cytoplasm is divided. In the laboratory, organism A can proliferate on
agarose supplemented with bacteria.
Dr. S cultured organism A derived from the soil in the NIG campus. Organism A can be either
Yellow (Y)-type or Green (G)-type. When Dr. S isolated a single Y-type individual and
proliferated it, she found that the descendant population consisted of mostly Y-types and a few
G-type individuals. On the other hand, when a single G-type individual was proliferated, the
descendant population contained mostly G-types and a few Y-type individuals. By observing
carefully, Dr. S found that conversions from Y-type to G-type and from G-type to Y-type occurred
at very low frequencies and only immediately after cell division.

Question 1 Dr. S wants to determine the conversion frequencies from Y-type to G-type
and from G-type to Y-type. If you were Dr. S, how would you measure the conversion
frequencies? Please explain your experimental design.
Question 2-1 Dr. S found conversion frequencies of 1% per cell division for Y-type to G-type
and 2% per cell division for G-type to Y-type, (both were measured at 15˚C). When Dr. S cultured
organism A for a long time, the ratio of the numbers of Y-type and G-type individuals became
stable. Given that Y-type and G-type individuals proliferate at the same speed (i.e., have the
same number of cell divisions per unit time), please predict the steady-state ratio of the
numbers of Y-type and G-type cells. Please describe the calculation process and explain the logic
behind your calculation.
Question 2-2 When the population of organism A described in Question 2-1 was
transferred to 30˚C and cultured for a long time, most of the individuals became G-type. A
possible explanation for this phenomenon would be that the temperature shift (15˚C to 30˚C)
altered the conversion frequency (i.e., increase of Y to G conversion frequency and/or decrease of
G to Y conversion frequency). Please propose alternative scenarios to explain this observation.
Please describe multiple possibilities, if it is possible.

Question 3-1 Dr. S is interested in the mechanism by which organism A reversibly converts
from Y-type to G-type and also from G-type to Y-type. Please propose a possible mechanism for
these conversions.
* The fact that A is multinuclear should be considered.
Question 3-2 Please design an experiment and/or analysis to test the hypothesis that you
described in your answer for Question 3-1.
Question 4 What else would you want to study if you had organism A? Please propose
two distinct projects (Projects 1 and 2). For each project, please describe your “question” (what
exactly do you want to know?) and “the outline of the research” (What kinds of experiments will
you perform to solve the question? What kinds of results do you expect? etc.).
