Diagnostic Agglutination TestingDiagnostic agglutination kits are used for the identifi cation of bacteria growing on culture media and for the direct detection of specifi c antibody produced due to microbial infection. These tests are based on particle agglutination with simple visual reading. The tests can be performed easily on single or batched samples. The provision of appropriate controls ensures the reliability of results.

As an example, the test reagent can be a suspension of sub-micron particles which have been sensitized with antibodies that react specifi cally with antigen from the target organism. When a portion of a colony or a sample containing target organism antigens is emulsifi ed with the test reagent, the antibody present on the latex, blood cell or carbon particle surface, binds to the homologous antigens. This binding causes considerable cross-linking of the particles to produce large clumps. The clumps, when of a suffi cient size become visible to the naked eye during the rocking of the card. In addition to the presence of agglutinated clumps, the particles are removed from the liquid phase causing a noticeable, and often complete, clearing of the smooth background. In a negative reaction, when the organism does not

carry the homologous antigen, the antibody-coated particles do not bind to the bacterial antigens, no cross-linking occurs and the coloured suspension remains smooth.

Three particle types are used for the assays: stabilized sheep red blood cells, carbon particles and coloured latex particles – colours include blue, white, yellow, red and green latex. All of these formats give highly visible reactions when viewed against the supplied, high quality disposable reaction cards. Most of the tests are based upon antibody/antigen reactions, which are made visible by the presence of the coloured particles.

The tests give easily visible, rapid reactions and offer the benefi t of minimal sample preparation with assay times from only 20 seconds up to eight minutes.

The Dryspot range uses predispensed test and control latex dried onto reaction cards. This means there is less handling of reagents, no dispensing errors and less chance of contamination, resulting in improved reproducibility. This dried format has the added advantage of a 2 year shelf life and room temperature storage.
