Working from 2 years on various microbiological topics like bacteriological, viral and fungal infection. Bacteriology: I - Molecular detection of MRSA by mec A and Luk-PV genes in gram positive clinical isolates. II - Molecular diagnosis of metallo beta lactamases (MBL’s) genes like VIM, IMP, NDM-1 genes in gram negative clinical isolates and genotyping of NDM-1 gene was carried out by DNA sequencing. III- Molecular detection of ESBL & AmpC genes in ICU patients. Virology: I- Molecular diagnosis of HBV, HCV, HIV-1 viruses by PCR.

Fungus: I- Molecular diagnosis of Candida albicans by detecting ALS-1 & HWP-1 genes in HIV-1 clinical isolates. II- Effect of Silver Nano particles on ALS-1 & HWP-1gene expression in overnight cultured C. Albicans cells. “Molecular analysis of drug resistance in HIV-1 infected patients in North India”

The main aim of the proposed study was to identify the drug resistance associated mutations in HIV-1 protease and reverse transcriptase region of polymerase gene among North Indian population and assess the phylogenetic relatedness of these patients to known HIV-1 strains. My Ph.D. thesis research work involved the PBMC extraction from whole blood, pro-viral DNA extraction, PCR for Protease and Reverse transcriptase gene of HIV-1genome, DNA sequencing of amplified products, DNA sequence analysis by MEGA, version 4.0 and Bio Edit version 7.0.9.0 software, genotypic drug resistance interpretation was done by different HIV algorithms and finally HIV-1 genotyping was done by REGA v2.0 and constructed the phylogenetic tree by MEGA v4 software.

“Occult hepatitis B virus infection: diagnosis, immunological profile and clinical implication in patients with chronic liver disease”

Amplified PCR products of particular gene regions of HBV were sequenced to find out the HBV genotypes circulating in this region and detected mutations in pre-core gene of HBV. We quantified the Th-1 and Th-2 cytokines by using ELISA and correlated with the clinical outcomes. We also detected the SEN and TTV viruses by using PCR in patients who were serologically negative for HBV and HCV.

“Effect of Taurine chloramine on heme oxygenase-1 expression in murine macrophages (Raw 264.7 cell line)”

Heme oxygenase-1 is a stress response & antioxidant enzyme. To know the effect of Taurine Chloramine on HO-1 activity in macrophages, We were studying mRNA expression of HO-1 gene, iNOS gene and some other genes, protein expression (by Western blot) of HO-1 & other proteins, Nuclear translocation of NrF2 transcriptional factor and NrF2 & ARE binding by EMSA (Electrophoretic Mobility Shift Assay). Finally, we wanted to know that what specific mechanism involves in Heme oxygenase-1 induction in macrophages by Taurine Chloramine.

“Molecular subtyping of HIV isolates by partial nucleotide sequencing of integrase gene”

Amplified products of HIV-1 Integrase gene were sequenced. Nucleotide sequences were edited & phylogenetic trees were constructed for molecular subtyping of HIV isolates. Study revealed majority of isolates to be of HIV-I Subtype C.
