1. Plasmid encoded traits in E. coli of man and animals ( Doctoral work)

One hundred ninety four strains of E. coli of different clinical conditions of man and animals were tested for their antibiotic resistance and virulence factors such as enterotoxin, haemolysin, colicin, adhering factors. Compatibility of drug resistance with virulence factors was determined. Findings indicated a close relationship between drug resistance with enterotoxins production as well as colicin production. Transferable R-plasmids have been detected by conjugation and plasmid curing experiments. Co-transfer of antibiotic resistance with virulence factors have been recorded in many E. coli strains.

2. Impact of wastewater on crop productivity and soil health(1996-2002 IOC,

Project) In a collaborative study with department of Botany, AMU and Mathura Oil refinery, Mathura India, the investigator has evaluated long term impact of Mathura Oil refinery treated waste water on physiochemical and microbiological characteristics of soil, irrigational quality of waste water, heavy metal content and accumulation in soil and crop produce. The possible impact of wastewater application on emergence of metal resistance was recorded. Treated wastewater of Mathura oil refinery was found suitable for irrigation purpose for crops like wheat, mustard, Arhar, vegetables and sugarcane. Continuous application of wastewater resulted in slow increase in heavy metal content of field soil but is under permissible limit. No significant amount of heavy metal accumulation was detected in crop produce. Microbial dynamic of soil was not adversely affected. Soil bacteria showed high level of metal resistance in vitro and in soil. Conclusions: Application of wastewater for crop irrigation may be an economical way of reutilization in agriculture however continuous monitoring on wastewater irrigational quality is needed. Impact of long term use of wastewater on soil health and quality of crop produce should be evaluated regularly.

3. Performance of biofertilizers and characterization & development of indigenous

strains:(Departmental projects and IOC project with Botany Department) Performance of biofertizer preparations (obtained from IARI, New Delhi) were evaluated in crops like Moong, Arhar, Chick pea, Wheat and Sugarcane at Aligarh and Mathura. Performance of Rhizobium in Arhar is on the expected line however it is not consistent in Moong. Azotobacter inoculation in wheat gave significant increase in yield. On the other hand sugarcane variety (Local Harirani) responded significantly to combined inoculation of Azotobacter + Azospirillum. Inoculation combination, Azotobacter + VAM showed antagonistic response. Indigenous diazotrophic bacteria like Rhizobium, Azotobacter and Acetobacter diazotrophicus (Gluconoacetobacter daizotrophicus) have been isolated from different soil and crops. Many of these potential inoculants strains are identified and characterized for their plant growth promoting activities like production of plant growth regulators, nitrogen fixing ability, solubilisation of phosphate and biocontrol activities.

4. Metal -Microbe Interactions :

Metal tolerance among diazotrophic bacteria, PGPR and plant growth promoting fungi (PGPF) are isolated from agricultural soils. Effect of metals on bacterial

survival and legume-Rhizobium-symbiosis showed advwerse effects at elevated metal concentrations. Metal-fungi interactions: metal tolerance, effect of metal on spore germination, biomass production, phosphate solubilisation, production of acetic acids, siderophores and extracellular enzymatic activity and inoculation response to crop yield have been evaluated. Metal biosorption potential of soil fungi and identified fungi could be exploited in removing heavy metals from aqueous solutions.

5. Bioactivities of Indian medicinal Plants, essential oils and natural products): In search of potential bioactive compounds from traditionally used Indian medicinal plants so far more than 120 medicinal plants have been screened against multidrug resistant human pathogenic bacteria, yeast and several phytopathogenic fungi. On the basis of primary screening various plants have been identified as potential medicinal plants to be exploited in the management of Infectious diseases of human, animal and plants caused by bacteria and fungi. Phytochemical analysis by TLC and TLC- bioautography revealed major active compounds like tannins, flavanoids, alkaloids, saponins, steroids, napthoquinone and glycosides in different plant extracts. Activities against multidrug resistant ESBL producing Enteric bacteria, R-plasmid harbouring bacteria, Methicillin resistant S. aureus (MRSA) have been determined in vitro and active compounds are under investigation In vitro antioxidant activities and antimutagenic activity of medicinal plants are under investigation from identified plants including spices. On the other hand potential plant extracts are under investigation for development of standardized herbal extracts for therapeutic potential in animal models are needed.

6. Antifungal, antivirulence and antibiofilm activit ies of essential oils (ICMR-SRF Research projects)

Antifungal activities against drug resistant and sensitive Candida albicans and filamentous fungi like dermatophytes and Aspergillus spp have been reported. Anti-virulence and anti-biofilm properties of essential oils and their active compounds were screened and their possible mode of action on biofilm and virulence factors at sub MICs has been proposed.

7. Bacterial Quorum sensing interference by natural products A novel anti

Infective drug target) ( UGC Project and Departmental Project work) Bacterial quorum sensing signal molecule (AHLs) production and detection assays were standardized and used for screening of soil microbes for their ability to modulate quorum sensing regulated functions in Chromobacterium violaceum and Pseudomonas aeruginosa. Quorum sensing quenching bacterial isolate was isolated from soil. Screening of medicinal and food plant extracts and plant essential oils are under study for their possible interference with bacterial quorum sensing. Some of the plant extracts were identified as potential source of anti-QS compounds. Effects of natural products on QS regulated functions and other non QS regulated virulence factors of MRSA (S. aureus) and E. coli are also under investigation.
