The COVID-19 pandemic has brought the world literally to a standstill. There is still no cure or a vaccine for this highly

infectious disease. The WHO declared COVID-19 as a pandemic on 11 March 2020 and ever since, scientists all over the world have been in pursuit of a cure or a vaccine and have been developing technologies to combat this pandemic.

CSIR, through its 38 laboratories and expertise in diverse areas ranging from aerospace to buildings to genomics to chemicals, quickly sprang into action developing COVID-19 interventions. For effective planning and strategizing, CSIR swiftly set up five COVID-19 verticals including disease surveillance, drugs & vaccines, testing & diagnostics, PPEs and supply chain management.

CSIR's rich R&D knowledgebase is embodied in the rapid testing, cost-effective diagnostic kits, virus culture, genome sequencing and disease surveillance. With the perfect blend of scientific knowledge, technical expertise, industry linkages and governmental interfaces, in about 100 days CSIR has come up with almost as many technologies and products to be deployed against COVID-19. From innovations and improvisations, these technologies have different levels of sophistication. Right from ventilators to indigenous diagnostic kits to clinical trials of repurposed drugs to much needed WHO stipulated hand sanitizers and masks, CSIR has been at the forefront in the nation's crusade against COVID-19. CSIR has developed direct RT-PCR method and novel FELUDA diagnostic kit for COVID-19. When the country faced a shortage of PPEs, CSIR developed a protective coverall and today coveralls are being manufactured in the thousands. CSIR developed a ventilator in a record 36 days. Even the mundane but must-adorn mask underwent various kinds of innovations in terms of the materials woven or the coatings layered in them.

About 60 out of the 100 or so technologies listed in this compendium have already been transferred to industry partners. The COVID-19 technologies are licensed on a non-exclusive basis and it is heart-warming that all our industrial partners have been quick to respond and engage with us in the larger interest of taking these technologies to the needy in a short time.

With the lockdown effecting reverse migration, CSIR took upon itself to provide tons of nutritious and immunity-boosting foods. The recently launched KisanSabha portal and App supports the farmers to get better prices for their produce. It is very heartening that the farming community has received the App very well.

We hope that this compendium will not just give an insight into the COVID-19 technologies developed by CSIR so far, but will also spur more innovations. An 'Aatma Nirbhar Bharat' is high on our agenda, and we are eager to partner with more industries to take these technologies to the users. Your feedback and ideas are very welcome.

The ongoing pandemic of Coronavirus Disease 2019 (COVID-19) is caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS CoV 2). The World Health Organization (WHO) declared the outbreak to be a Public Health Emergency of International Concern on 30th January 2020 and recognized it as a pandemic on 11 March 2020. As of 1 July 2020, there are 5,86,956 cases of COVID-19 in India.

Developing technological solutions that are needed to tackle this unprecedented challenge requires strong and cutting edge R&D capability and capacity in diverse disciplines ranging from biological sciences (vaccines, diagnostics, sequencing, testing of drugs) to chemical sciences (drugs: repurposed and new drugs) and engineering (ventilators, PPEs) and interdisciplinary approach transcending barriers.

CSIR is uniquely placed as an S&T organization to deliver these at this critical juncture, having proven expertise in sequencing (first human genome sequencing), diagnostics for rare genetic diseases and being the fountain head of generic industry in the country. With expertise ranging from development of aircrafts to advanced materials, CSIR is committed to harness its vast scientific human resource and its considerable capacity and is well poised to serve the diverse needs required immediately and also plan for long term solutions that are needed to control this epidemic. This calls for a strategic, well-coordinated and integrated approach in cooperation with all relevant stakeholders and aligned to government’s strategy.

CSIR Strategy to Fight COVID-19

CSIR has advantageously positioned itself to pursue the focused R&D to develop, integrate, scale-up, and deploy necessary technological interventions for combating coronavirus pandemic in the country. Considering the multifarious problems created by coronavirus which require interventions in several areas and multi-pronged strategy, CSIR has set up five technology verticals for addressing the pandemic.

These verticals are need-based and span multiple research labs and disciplines and draw upon the strength of scientists, students and harness it for the fight against COVID-19. In addition, CSIR is also working on promoting rural employment and providing ready-to-eat food to the needy migrants.

CSIR has developed several COVID-19 focused technologies and products ranging from sanitizers & masks to

improved tests and diagnostics and ventilators, buildings and structures. The technologies, products and processes listed in the compendium are available for non-exclusive technology transfer. Out of 105 technologies and products in this compendium, 58 have been transferred to various industry partners. For the others, the technologies are in different stages of readiness, the most basic being a working prototype. Interested start-ups and industries may contact the concerned CSIR laboratory for further discussion, and technology transfer.

The key to flattening the curve in the number of Coronavirus cases is the detection of the infected at the earliest and isolating them. A combination of digital and molecular surveillance with rapid diagnosis is the need and CSIR is striving towards that using multiple strategies. CSIR was one of the first institutions to carry out testing in the country. With state-of the-art genomics and molecular biology laboratories in the CSIR network, CSIR has not only been at the forefront of testing but also developing cost-effective and accurate diagnostic kits for detecting the COVID-19 virus.

Some of the technologies developed by CSIR are:

Dry Swab-based RNA Extraction Free Direct RT-PCR Diagnostics

► Dry swabs (with nasopharyngeal samples on it) are sent from collection centres to testing centres.

► TE buffer is used to isolate the biological samples, skip RNA isolation, and proceed with RT-qPCR without compromising the sensitivity of the COVID-19 tests.

FnCas9 Editor Linked Uniform Detection Assay (FELUDA)

► Detection of pathogenic sequences or variants in DNA and RNA through a point-of-care diagnostic approach is useful for rapid clinical prognosis.

► FnCas9 Editor Linked Uniform Detection Assay (FELUDA) employs an accurate enzymatic read out for detecting nucleotide sequences, identifying nucleobase identity and inferring zygosity with precision.

► FELUDA output can be adapted to multiple signal detection platforms and can be quickly designed and deployed for versatile applications including rapid diagnosis of infectious diseases such as COVID-19.

► The knowledgebase encompasses a comprehensive pipeline including protein expression, sgRNA design, preparation of RNA complexes, detection methods that are dependent on applications, SOPs for pathogen detections and SNP detections.

►► Consists of a disposable micro fabricated amplification chip, capable of handling 20-150 µl sample volume.

RT-LAMP Diagnosis of COVID-19►► Sample is collected as a nasopharyngeal swab in a viral transport medium.

►► RNA is extracted from the sample and used for RT-LAMP.

►► RT-LAMP based diagnosis relies on conversion of RNA to cDNA and then an isothermal amplification of specific viral regions through specially designed primers.

►► Addition of SYBR Green I dye to the reaction mixture results in green fluorescence on accumulation of amplified DNA in a positive sample. Thus a simple visual detection can ascertain if the sample is COVID-19 positive or negative.

►► This rest is simple and does not require expensive instrumentation and expertise.

►► The sensitivity and accuracy of the test almost matches the current gold standard qPCR test.

One-step RT PCR Kit-COVID-19►► Recombinant clones for highly efficient Reverse Transcriptase and hot start Taq DNA

polymerase.

►► Formulated one step picomolar sensitive RT PCR kit.

►► Tested efficiency of the kit against COVID-19 samples using TaqMan probes.

To combat the virus, CSIR is exploring all avenues and supporting new ideas that have a clear deployment strategy. CSIR has further defined its strategy in this vertical by setting up focused groups to look at multiple pathways for developing drugs:

i) Molecular modelling of drugs; ii) Host-target interactions; iii) Crystal engineering;

iv) Batch to continuous process; v) In-silico screening of existing drugs and natural products;

vi) Scouting for non-infringing routes, and vii) Host-dependent pathways for APIs.

Process Development of Drugs Repurposed for COVID-19►► Process developed for Nitazoxanide, EIDD 1931 & EIDD 2801.

►► Non-infringing routes developed for the APIs.

►► Nitazoxanide is currently under clinical trials in Mexico.

Coronavirus Culture ►► Stable cultures of coronavirus in

Process for Niclosamide►► The gram scale process for synthesis of

niclosamide developed.

►► It is an FDA approved anti-helminthic drug. It has been found to be a potential anti-viral agent and is one of the top 25 shortlisted repurposed drugs.

►► The synthetic process technology has been developed.

Repurposing of Colchicine for COVID-19 treatment►► This is a FDA approved drug for gout.

►► Tablets are available with the Cadila.

►► The same dose of tablets (0.5 mg) is propsoed to be used for COVID-19 trial.

►► The clinical trial strategy for conducting Phase II clinical trial of colchicine along with a comparative arm of SoC, has been prepared.

Synthetic Process for Camostat Mesylate►► Synthesis of Camostat mesylate, starting

from indigenous and less expensive raw materials that are mostly available in India.

►► The innovation includes the improvement of existing methodologies by reducing the number of steps.

been developed using indigenously available key starting materials.

►► CSIR-CDRI has received the permission from Central Drugs Standard Control Organization for conducting the Phase III clinical trial with Umifenovir entitled "Randomised, double blind, placebo controlled trial of efficacy, safety and tolerability of Antiviral drug Umifenovir vs standard care of therapy in non-severe COVID-19 patients.”

►► Tilorone is used as a broad-spectrum antiviral discovered more and chemical process that is an improvement of existing synthetic strategy for Tilorone dihydrochloride using less expensive and easily available raw materials.

Zinc Gluconate Vitamin C Formulation (Zincona-C) for Immunity Improvement►► The Formulation is based on green

technology for Zinc gluconate and natural vitamin C i.e. Acerola Cherry extract.

►► Capsules and syrup have been prepared as per RDA.

►► The product is being taken forward for the clinical trials.

►► Applied for trade mark (Zincona-C).

Hospital Assistive Devices and Personal Protective Equipment (PPE)A major challenge during the corona pandemic globally has been the shortage of hospital assistive devices such as ventilators and oxygen enrichment devices. Another critical requirement in this pandemic has been the need for personal protective equipment (PPEs) to the caregivers who include doctors, nurses and para medical staff given the infectious nature of the virus.

Under this vertical, CSIR has developed Hospital Assistive Devices (HADs) such as ventilators, oxygen enrichment units, hands-free dispensers, electrostatic disinfector, and several kinds of personal protective equipment.

Not just engineered devices and equipment but relatively simple but essential items like hand-sanitizers and face masks have also been developed by many labs and distributed widely as a social responsibility of the CSIR labs and institutes.

Oxygen Enrichment Unit (OEU) based on Membrane Technology For Oxygen Therapy►► A startup of CSIR-NCL, Genrich Membranes

has developed Oxygen Enrichment Unit (OEU) based on innovative, indigenously developed Hollow-Fiber Membrane technology. This unit provides ~35-40% Oxygen enriched air.

►► The device consists of (1) membrane cartridge (filter), (2) pressure-gauge, (3) flowmeter, (4) humidifier bottle, (5) nasal-cannula, (6) oil free compressor and (7) tubing & fittings.

►► The atmospheric air under certain pressure (4-5 bar) passes through membrane cartridge wherein oxygen is permeated through membrane surface preferentially over other gas components in the air. This happens due to intrinsic high sorption capacity of oxygen in to membrane matrix.

►► Double side polished silicon wafers with (100) orientation and a thickness of 370 ± 1 µm are used as starting substrate. Thermal oxide used as insulator and etching performed using buffered HF. Poly silicon resistors are implemented on a diaphragm achieved by back-side etching. Poly resistors are connected in wheatstone bridge configuration. A commercial interface IC interfaced with a closed bridge pressure sensor. An on chip temperature sensor compensates temperature induced non-linearities generated from sensor.

Prefab Foldable Modular Structure for Makeshift Transit Hospital for COVID-19: Poli-Tal(M)►► The modular unit is a single foldable unit

having a total weight of around one tonne. This structure module can be folded flat so that it can be transported easily. The module can be fabricated in a yard with simple fabrication technology that utilizes welded standard steel sections. The fully folded modules can be taken to the site and deployed after making the site levelled and ready.

►► One hospital of around 100 beds can be installed within a week including beds,

electricity and water supply etc. All piping can be accommodated within the panels including switch boards and pipe connections.

►► The modular units are spacious and are multifunctional, can be used for patients, doctor's cabins and testing labs.

►► The total size of the full size module is around 6m x 3m x 0.150m which can be easily transportable from one place to another. The size is customizable during fabrication as per the requirement.

CSIR-CSMCRI Disinfection chamber for PPEs has been developed which aids in the disinfection of face mask, face shields.

CSIR-CECRI A process for the electrochemical preparation of sodium hypochlorite (1.0 % NaOCl) disinfectant from common salt has been developed. The solution is mainly used as a disinfectant in public places and hospitals to kill bacteria, fungi and virus. This is green technology with no chemical effluent for disposal.

CSIR-CEERI Multi-wavelength UV Source for Killing Pathogens in Real Time (MUKT) is a plasma-UV based electronic easy-to-use pathogen inactivation system designed for its multi-purpose and diverse applications in medical, domestic, food and grocery stores. There are two versions of MUKT, namely MUKT Portable Version and MUKT Handheld Version. The systems are easy to operate with normal AC power supply.

CSIR-CMERI A pneumatically operated mobile indoor disinfection (POMID) unit has been developed which can be used for deactivation or killing of pathogenic microorganisms present inside a chamber like rooms, halls, building corridors spring mist of water having chemical disinfectants

CSIR-CMERI A portable touch free soap-cum-water dispensing system - compact version has been devleoped. When a person approaches the dispenser, a predefined amount of soap (approx. 5ml) will be dispensed from the tap After rubbing his/her hands for twenty (20) seconds, clean water is dispensed from the same tap.

CSIR-IMMT The touch-less hand sanitizer is a contactless machine that uses a low cost sensor and motor system to push and release the dispenser top. When the hand is brought nearer to the dispenser, the required amount of liquid is falls on the palm.

Hand SanitizersEven as the pandemic made its appearance, it was realized that preventing the spread of the highly infectious virus is essential. Social distancing, washing hands frequently and using hand-sanitizers have been very widely advocated. This led to a heavy demand for hand-sanitizers which in turn caused scarcity of the hand-sanitizers.

Many CSIR laboratories and institutes stepped in and in accordance with the WHO guidelines, thousands of litres of much-needed lab-scale hand sanitizers were prepared and distributed to not only its own staff but also to the local administration, health-care professionals, police personnel and so on.

Many labs also innovated on the formulations by augmenting it with moisturizing agents and fortifying it herbal and aromatic compounds. Many labs have transferred their improved processes to industry partners.

CSIR-CMERI A tractor mounted road ranitizing unit has been developed that can be deployed in long stretches of highways, vicinity of toll plazas etc. where there is a massive volume of traffic and good chance of infection spreading.

CSIR-IMMT The UV Disinfectant has been developed with 125 W UV bulb and a rotating base fitted with a shutter door to disinfect personal belongings like masks, key rings, currencies, hanky, tissue, papers, bills etc. and other objects.

CSIR-CEERI UV-C based sanitizer system has been developed for disinfecting fomites and other surfaces. It has a uniquely design has the UV-C lamps maintains uniform intensity throughout the chamber. The system is capable of disinfecting the exposed objects in ~2 minutes. It is equipped with safety switch to ensure automatic switching off of lamp in case door is opened during operation to avoid UV exposure risk.

In addition to the five verticals that have produced several technologies to manage the pandemic through R&D interventions, CSIR has also focussed on the food technologies. The lockdown had caused large-scale reverse-migration all over India with people travelling enmasse to their hometowns. To overcome the non-availability of food, CSIR stepped in to supply food to the needy Further, immunity boosing food and food supplements were also formulated and supplied.
