FREEZE CONTROL® BIOFREEZER TECHNOLOGY

USER-PROGRAMMABLE MODELS (CL3300, CL8800)

CryoGenesis™ Version 4 or 5 temperature control software is included with user-programmable models. This Windows™-based software allows the user to design temperature protocols by generating a set of ramps (including target temperatures, rates, holds, and alarms); manage protocol libraries (save, load, edit); execute protocols (use real time data, allow user interactions); log progress and allow for clinical validation documentation. As with preprogrammed models, final program state may be “Hold” or “Freefall” to the temperature of LN2.

PREPROGRAMMED MODELS (CL2200, CL5500, CL8800)

The customer may specify target temperatures, rates, and holds on preprogrammed temperature controller models. Sets of programs are written and stored onto a single memory “chip”, which is easily user-replaceable. Final program state can be “Hold temperature at finish” or “Freefall at finish”. If the customer requires future program changes, a replacement program chip can be provided. Preprogrammed mode offers a level of increased security, because the programs cannot be inadvertently deleted or modified.

Biogenics offers a complete line of FREEZE CONTROL Biofreezer systems, precision engineered for cryopreservation of biological specimens. FREEZE CONTROL systems provide a patented and reliable method for heat transfer and temperature regulation in the freezing of biological material for long-term preservation and viable recovery. These controlled-rate, non-pressurized liquid nitrogen systems include a FREEZE CONTROL temperature controller, a cryo- chamber and a cryobath. Options in system configurations are available—Biogenics will work with you to determine the most appropriate combination for your specific application requirements. Four models of preprogrammed and/or user-programmed temperature controllers are available.

There are five easy steps for using FREEZE CONTROL biofreezer systems: 1. Switch on the controller. 2. Stand the cryochamber in the LN2 cryobath. 3. Load specimens when the chamber has reached the “start” temperature. 4. Select and execute the desired temperature program. 5. After a run, load the next batch of specimens and continue.

HOW FREEZE CONTROL SYSTEMS WORK

There are three essential components to every FREEZE CONTROL system: controller, cryochamber, and cryobath. The cryochamber stands directly in a cryobath of liquid nitrogen and is connected to the temperature controller. The controller regulates the temperature of the specimens precisely and reliably. No special installation is required—the user can quickly set up the system, use it and store it away.

• SAFETY Only small amounts of liquid nitrogen are required. There are no pressurized LN2 vessels, and no alcohol or other volatile, toxic or flammable liquids are used.

• PRECISION TEMPERATURE CONTROL The rate of cooling and thawing is variable, according to user requirements.

Temperatures can be held constant at any point within the control range. Controllers can also be used to provide controlled-rate specimen thawing. Thermometry and temperature control circuits are calibrated to accuracies of better than ± 0.1°C.

• ACCURACY The temperature can be precisely specified and accurately maintained at all times. The uniformity of temperature along and between straws or cryovials also ensures greater consistency and greater success.

• VERSATILITY With models using CryoGenesis™, practitioners can design and use an unlimited number of temperature programs of varying complexity, with up to 50 ramps and unlimited duration. A signal tone—from the controller, computer, or both can be included in the program to alert the operator when intervention is required, for example, at “seeding” temperature and/or at program finish. In the CL8800 model, up to 16 programs can be preinstalled to allow the unit to operate without a computer; yet the temperature datalogging facility in CryoGenesis is still active, so temperature cycles can be validated even in preprogrammed mode of operation.

• RELIABILITY The freezers use only solid state integrated circuitry. There are no compressors, solenoids, fans or valves with moving parts to inevitably wear out. The freezers are vibration free and noiseless.

• EFFICIENCY AND ECONOMY Consumption of LN2 is less than 1 liter per hour. The low power consumption, typically less than 60 watts, makes fail-safe battery power practical. There are no other consumables. Proprietary circuit design and component selection ensure that calibrations have long term stability, contributing to low maintenance and reduced service costs.

PRODUCT CODE: CL8800M The FREEZE CONTROL CL8800 system is a user programmable/preprogrammed biofreezer incorporating a controller with temperature acquisition and datalogging capability. Combined with CryoGenesis V5 software for Windows, a CC23S Straw Cryochamber and either the LNS, LNL, or DX2 Cryobath, this system is perfect for use in a busy lab for routine or research applications. When connected to a computer, the CL8800 provides a facility for full validation of actual freezing cycles that can be assigned to specific file names. The CL8800 is fully user-programmable when run from any IBM compatible PC operating under Windows 3.1 or higher, using our Windows software, CryoGenesis V5. The CL8800 can also be preprogrammed to allow the system to operate without a computer. Up to 16 user-specified programs can be preinstalled. When connected to a PC, the CL8800’s built-in temperature acquisition facility allows CryoGenesis to datalog actual specimen temperature. The temperature control range is +40°C to -120°C. Validation of cryopreservation process can also be performed via an analog chart recorder, using the included temperature output jack. The FREEZE CONTROL® CL5500 system is a preprogrammed biofreezer incorporating a temperature controller with built-in rechargeable battery for power failure protection or portable operation. This controller also can be powered from an external 12-volt source. The 5500 offers complete peace of mind when operating in areas of questionable AC mains power.

The CL5500 system comes with eight standard or custom resident programs. The CL5500 system is ideal for routine lab applications where the range of controlled temperature will not exceed -43°C. This system is also perfect for the mobile embryo transfer practitioner. For this application, a custom form- fitted transport case built to withstand the rigors of road or air travel is available.

This compact system, with its built-in power pack, offers failsafe protection against power failures or blackouts. The specimens being cryopreserved will never be affected by such incidents. From a full charge, running time is 3 to 4 hours (continuous) in absence of AC mains power. The temperature control range for the CL5500 is +20°C to -43°C. FREEZE CONTROL cryochambers are unique. Their patented design ensures symmetrical heat transfer from all specimens to liquid nitrogen. The highly conductive material used for the chamber ensures a very high degree of temperature uniformity of each specimen whether Liquid Nitrogen levels are high or low. Specimen temperature is monitored continuously and is measured by a precision platinum temperature sensor permanently mounted in the core. Specimens can be safely accessed for manual seeding, without exposing them to temperature fluctuations. The conductive core of the chamber, and the tight thermal coupling between specimens and core, ensure that latent heat is efficiently removed during the nucleation process.

All cryochambers are available in two styles: STANDARD or FAST, designated by product code suffixes “S” or “F”, respectively. The STANDARD design is suitable for most applications, whereas the FAST models are appropriate for applications where a higher- speed cooling rate is desired. We are pleased to guide you in the selection of cryochambers to match your requirements.

The Standard CC23S Straw Cryochamber is included with all biofreezer systems unless the customer order states otherwise.

Biogenics FREEZE CONTROL biofreezer systems consist of a temperature controller, cryochamber, and cryobath. A range of FREEZE CONTROL cryochambers is available to accommodate different specimen containers (e.g., straws and/or cryovials, security straws, etc.), with cooling rates appropriate for virtually any protocol. There are two cryobath sizes available, interchangeable according to customer preference.

VHC™ CRYOVIAL / HIGH CAPACITY STRAW CRYOCHAMBERSBiogenics offers several options in VHC cryochambers for use with any of our biofreezer systems. Biogenics Service Contract increases the economy, convenience, and efficiency of servicing your laboratory cryosystem.

Annual calibration and servicing of all Freeze Control® biofreezers and incubators is recommended to ensure continued accurate and trouble-free operation and validation of the equipment. Biogenics is the only company in North America authorized to service Freeze Control.

Biofreezer servicing includes a 15-point quality assurance protocol which eliminates any possible “drift” in previously calibrated settings, scrutinizes your cryochamber for the development of any abnormal temperature gradients, and allows us to compare the values of previous settings to evaluate any performance degradation.

Further, diligent QA is mandated by CAP, and is a general part of the overall CLIA guidelines. Inspectors routinely require documentation that such service is being maintained.

The Biogenics Service Contract provides the above for a term of five years, plus the following:

With the CVM1 system your specimens and the vitrification media never touch liquid nitrogen during vitrification or in storage thereafter. This eliminates exposure of the specimens to undesirable elements, viruses, fungi, and other agents, which may be present in liquid nitrogen.

Brilliantly designed with the aspect of repeated production in mind, the system consists of a sterilizable metal block which is comprised of a proprietary alloy and coating engineered for maximum cooling rate and specimen protection.

Complementary aspects of the system are the unique, proprietary fibre-plugs and their accompanying protective sheaths. The small hook on the end of the fibre-plug is designed to effectively suspend a micro-droplet of vitrification media containing the specimen (eg embryo or ocyte).

BIOTHERM MSP

The BioTherm MSP is a microscope warm stage with a selectable temperature range of +25ºC – +50ºC (controllable temperatures start at 2ºC above ambient).

The warm stage plate has a very low profile (less than 6mm) making it suitable for a wide range of microscopes. A large 22mm aperture stage is available for use with inverted and stereo microscopes.

The BioTherm MSP can be used with Petrie dishes or slides.

Heating is applied uniformly across the whole plate, eliminating any hot or cold spots on the plate. BIOTHERM INC-12V/IRB-1 PORTABLE INCUBATORS/CELL CARRIERSThe BioTherm INC-12V and IRB-1 are portable incubators designed for maintaining live cell cultures, gametes and tissues at appropriate metabolic temperatures, with less than 0.05°C temperature fluctuation or variation between specimens. CRYOLERT™ LN2 LEVEL SENSING TANK ALARMS

Biogenics developed CryoLert in response to demand for an economical, true level-reading tank alarm systems. Research shows that to provide true protection to valuable biological specimens, an alarm should monitor cryogenic refrigerant level rather than tank temperature. The CryoLert continuously monitors actual liquid nitrogen level and allows the practitioner to specify an alarm point based on canister exposure out of LN2.

CryoLert liquid nitrogen alarm alerts the practitioner both audibly and visibly when LN2 levels fall below a pre-set point of safety or if the sensor or power has failed. LN2 levels routinely decrease due to slow evaporation or deteriorated vessel vacuums. Often, these levels drop rapidly without warning. The CryoLert was engineered with many features, including power failure indicator with auto reset, relay contacts for an external alarm, and an LN2 fill timer which disables alarm outputs thus preventing false alarms while filling the vessel or manipulating specimens.

BIOGENICS 6-WELL VENTED CULTURE DISH

Biogenics 6-well vented culture dish reflects an innovative rethinking of the specific requirements of the assisted reproductive laboratory. The features of this multi-welled dish were designed in response to the demands associated with handling, sorting and culturing spherical cell structures and other sensitive biological specimens.

Unique to the 6-well vented culture dish are its radiused corners in each well which allow specimens to “roll” into the center of each well and avoid sticking into corners–because the wells have no corners.

The vented lid allows for two positions indicated by molded keyways positioned 90º apart. The open position elevates the lid uniformly by 1mm allowing for gas exchange and prevention of contamination when placed inside an incubator, whereas the closed position allows for secure transport across the lab.

The center well’s patent-pending convex/concave geometry creates an annular “canyon” where specimens may be sorted prior to culturing. Alternatively, this well is useful for humidification.

Tight tolerance between the skirt edge and the well bottoms ensure good thermal transfer when the dish is place upon a heated stage.

Ribs on the dish lid interface with a mechanical interference fit on the skirt of another culture dish, allowing the dishes to be stacked vertically with reduced risk of spillage during transport or incubation.

Each well is numbered for the purpose of tracking individual specimens.

The additional two wells (compared to a 4-well dish) result in better economy and value.

The dishes are manufactured from 100% virgin, crystalline medical grade polystyrene.

All production lots of the dish are subjected to a rigorous 72-hour optical polish process, which results in a clarity surpassing all competitive products. After sterilization by gamma irradiation they undergo a 96-hour mouse embryo assay test wherein test results are typically 99 – 100% post culture.

Dishes are supplied in case lots of 120 dishes. Each dish is individually wrapped and sterile, thus reducing the potential waste of unused dishes.
