RNA interference
• RNA interference, known as RNAi, has expanded dramatically in a short-term period since its first discovery in 1990s. Several types of small silencing RNAs have now been discovered, including small interfering RNAs (siRNAs), microRNAs (miRNAs) and PIWI-interacting RNAs (piRNAs). Small RNAs are involved in regulating gene expression in many ways, and RNAi is being harnessed by scientists for laboratory research and for the development of new therapies for disease.
The most interesting aspects of RNAi are the following:
• dsRNA, rather than single-stranded antisense RNA, is the interfering agent
• it is highly specific• it is remarkably potent (only a few dsRNA
molecules per cell are required for effective interference)
• the interfering activity (and presumably the dsRNA) can cause interference in cells and tissues far removed from the site of introduction
