Dry Storage Freezers

Why is dry storage important?

Safety. Storage in liquid nitrogen can make vials shrink. This may

cause liquid nitrogen to seep into the vials, which on rewarming,

expand and explode as nitrogen vaporizes inside the vials. Dry

storage eliminates this possibility. Also, the Isothermal design

provides added user safety by eliminating contact with or

splashing of liquid nitrogen.

Cross Contamination. Studies have shown that viral, bacterial

and fungal pathogens can survive after suspension in liquid

nitrogen. Infected samples can cross contaminate other samples

in the same liquid nitrogen tank. Dry storage eliminates the

possibility of cross contamination.

Temperature Uniformity and Liquid Level

In conventional (non CBS) vapor phase freezers, the

temperature directly under the lid can be reduced by raising the

level of liquid Nitrogen in the bottom.

Whilst improving the temperature uniformity (gradient), this

reduces the amount of space available for storing samples in

vapor. This also makes the freezer more expensive in terms of

the number of samples stored.

In this situation, ask the customer to find out the relationship

between temperature gradient and the level of liquid Nitrogen

in the freezer, since the specified temperature may depend

heavily on the liquid Nitrogen level.

In CBS Isothermal freezers all the internal volume is available

for sample storage - free from liquid Nitrogen.

Temperature Uniformity and Temperature Probe Location

Encourage the customer to check with potential suppliers

about the operating temperatures of their freezers and where

the temperature sensor is located.

Cold air sinks and therefore the best location for the

temperature sensor is at the top, above the samples. On the

CBS Isothermal freezers, the sensor is located directly below

the lid and above the top of the racks giving the customer

confidence that all samples are stored below the indicated

{display) temperature - see typical V-1500 mapping data

below.

If the sensor is positioned lower down in the freezer, the

displayed temperature will look good but can the customer

be sure of the temperature of samples higher up?

This study shows that as the liquid nitrogen in your CBS Isothermal

Freezer evaporates, the internal temperature at probe #1 will rise

slightly to -182C. As the re-fill process starts, the temperature at

probe #1 will return to the normal operating temperature in the -189C

range.

The temperature of the other 5 probes increases very little, ranging

between -186C to -196C. Therefore, 85% of the freezer storage

space remains below -186C and only the top15% of the space will be

slightly warmer in the -182C range.

This study provides temperature data that confirms samples stored in

a CBS Isothermal Freezer will be well below critical temperatures.

Your CBS Isothermal Freezer is pre set to re-fill when the liquid

nitrogen level reaches a low set point of 10” (25cm) and stop filling

when the liquid nitrogen reaches the high set point of 17” (43cm).

These filling set points will provide excellent operating parameters.

However, they can be changed to suit your specific requirements.

Temperature Uniformity and Thermal Conductivity

Customer: "Other freezers also offer -190°C vapor storage"

To compete with the Isothermal concept and provide greater

uniformity, other manufacturers use the thermal conductivity properties

of aluminum and install aluminum plates (frames and carousel walls)

inside the freezer. The aluminum conducts heat from the top of the

freezer, which promotes evaporation and the resulting Nitrogen vapor

rises to cool the top of the freezer.

This approach has two disadvantages;

• Firstly, the more aluminum in contact with liquid Nitrogen, the

higher the consumption will be. This is why it is important for the

customer to compare the loss rate of the actual models they are

considering.

• Secondly, the use of frames and carousels reduces the amount of

space inside the freezer for samples. In contrast, the open interior

of the CBS Isothermal freezer provides maximum storage capacity

without the restrictions of carousels etc.

Customer: " I prefer Narrow Neck Lids as they keep the cold in"

In an effort to reduce cold air loss and liquid Nitrogen consumption,

manufacturers have introduced models with narrow neck openings. Whilst the

concept appears attractive, feedback from many customers highlights some

practical issues with this design.

Firstly, access to and location of samples is made more difficult. In contrast,

the CBS Isothermal design with Nitrogen vapor vents helps to hold the cold air

inside the freezer. This means that full-width lids can be used giving the user

unrestricted access for easier sample location.

The following temperature mapping data shows how the temperature inside a

V-1500 increases on lid opening. Even after 30 minutes the air temperature at

the top of the V-1500 remains well below the critical glass transition

temperature of water.

Secondly, if anything is dropped or becomes jammed in the carousel system of

a freezer with a narrow neck lid, it becomes almost impossible to retrieve or

repair.

Liquid Nitrogen Consumption

Customer: “Other freezers use less liquid nitrogen”

The only way to measure and compare LN2 consumption of freezers is by

using the Static Evaporation Rate (also known as Normal Evaporation Rate).

This is an industry accepted method for measuring the LN2 consumption of an

empty freezer under standardized conditions. (A copy of the method is

available on request).

Many manufacturers make claims about lower LN2 consumption and running

costs but then don't support this with data in their sales material.

If LN2 consumption is an issue, we recommend that customers ask potential

suppliers for the Static Evaporation Rate (SER) for the freezers they are

considering so that like-for like comparisons can be made.

It is important that the customer asks for the SER of the specific model they are

considering, including built-in options. Some manufacturers only quote a SER

for the base model, however the actual SER could be much higher if the

customer orders optional aluminum frames or a carousel.

The amount of time required to fill a freezer with liquid

nitrogen will vary with model. Listed below are the

estimated fill times for each CBS Isothermal Freezer.

V1500 – approximately 40 minutes

V3000 – approximately 90 minutes

V5000 – approximately 90 minutes

V5000 EH – approximately 90 minutes

Initial Filling Details

These fill times are based on a conventional 180 liter

and 230 liter supply tanks operating at 22 psi.

Fill times may vary with the use of bulk tanks and

vacuum jacketed supply lines.
