Sectioning
Optical sectioning
o Change the focus, higher power
o Higher power means smaller depth of field
Light microscope
Need to use electron microscope for smaller one
o Scanning gives surface
o Transmission goes through
Resolution refers to the ability to see distinct points as distinct
points
Diffusion
o Flow from higher to lower concentrations
Rate – mass diffusing in time t.
Proportional to how much surface are it has and
how long it has to go
Prokaryote
o Archeae very different from bacteria
Used bacteria term because people didn’t know what
were archeae
Ex. #8 – prelab viewing angle
All cells have:
o Cell membrane
o Cytosol
o DNA – circular piece of DNA
Genomes are very small compared to eukaryotes
Very efficient in encoding stuff
o Ribosomes – makes proteins
Cell = 1 um cube
pH 7 – how many H are within cell?
Volume?
1 = 1 x 10-4 cm. Thus (1x10-4)3 = 1 x 10-12 = 1 x 10-12mL = 1 x 10-15L
Because 1 cm3 = 1mL =
Ex. 1 um = 1x10-4
Vibrio Isolation
Vibrio Streaking
Different
DNA
The bases are very hydrophobic so they stack on top of each other
Connected by hydrogen bonds
Antiparallel
Anneal primers
o Growth is always from the 3’ end
o If want to make copy of DNA, need to have primer that makes
a new 5’ end and continues to build to a 3’ prime end
o Always start to make a new strand from the old 3’ end
