• Chitin ist ein Poly-ß-1,4 –N-acetyl-Glucosamin (poly-GlcNac)
• Biologische Funktion: Festigkeit, Schutz u.a gegen Frassfeinde
• Vorkommen: Exoskellett von Insekten und Crustacea, Zellwände von Hefen
und Schimmelpilzen (inkl. höhere Pilze) ; bis zu 30-60 % Anteile)
• Chitin ist ein fibrilläres Material in biologischen Komposite Strukturen
(Ausnahme: Diatomea), vergesellschaftet mit Proteinen (Matrix),
Polyphenolen (Insekten), Mineralien (primär Ca-Carbonat / Calcit in
Crustacea)…
• Chitin ist in 12 Mol/l kalte HCl löslich oder in LiCl / Dimethylacetamid (analog
Cellulose), Chitosan solubilisiert in schwachen Säuren…
• Nachwachsender, bioabbaubarer Rohstoff: ca. 1000 Mrd. t Chitin-Synthese /

• >50% Deacetylierungsgrad (des Chitins)
• In 1%iger Essigsäure solubilisierbar (Chitin ist unlöslich)
• Viskosität: 200 – 2000 mPas
• kommerzielle Chitosan-Präparate i.d.R. >85% deacetyliert
• Molmassen : 100 kDa - 1000 kDa (komplexiert oft mit Säuren wie Essigsäure, Milchsäure)
• Positive Ladung in saurer Lösung:
• Chit-NH2 +H2O ⇌ Chit –NH3⊕ + H2O
• pKa –Wert = ca. 6,3
• Chitosan solubilisiert , wenn > 50% der Aminogruppen protonisiert vorliegen
• Löslichkeit stark abnehmend oberhalb von pH 6,0 -6,5
• Max. lösliche Konzentration ist abhängig vom Chitosan-Typ : i.d.R. 10-20 g / l Chitosan
• Chitosan ist filmbildend , daher geeignet für Gele und Coatings

Chitosan: Antimikrobielle Aktivität
• Abhängig von: Chitosan-Typ, Ziel-Organismus, Randbedingungen (pH-Wert etc.)
• Hefen>Schimmelpilze> Gram-positive Bakterien>Gram-negative Bakterien
• Sacch. cereviseae –Fermentation durch 3,6 mg / l Chitosan inhibiert
• Stopp des Wachstums von Fusarium solani durch 4 mg / l Chitosan
• Wirkung von Molmasse abhängig : < 10kDa Chitosan ist aktiver als natives Chitosan (degree of polymerization : min 7)
• Je höher der Deacetylierungsgrad umso wirksamer antimikrobielle Effekte…
• (analog dem Anstieg der Löslichkeit bzw. dem Anstieg der elektrischen Ladung)
• Saure pH-Werte verstärken die antimikrobielle Wirkung ( Hürden-Effekte)
• Hohe Temperatur ( z.B. +37 °C) wirksamer als niedrige (z.B. +4 °C)
• Matrix-Effekte (großer Einfluss):• Chitosan bindet viele Lebensmittel-
Bestandteile wie Alginate, Pektine, Proteine, anorg. Phosphate (Polyphosphate), anorg. Polyelektrolyte, kleine Elektrolyte wie NaCl, Ca-Salze, Na-Phosphate , ionische Emulgatoren (Lecithin, DAWE ), etc.
