Supplementary figureSupplementary Fig. 1 The alignment of AoHex1 and Hex homologs from nine

other fungi. Amino acids in the C terminus are more conserved that those in the N

terminus. The peroxisome target signal sequences, S-R-L, exist in the C termini of

seven Hex proteins, while AoHex1 and a Hex homolog from D. haptotyla instead had

FKL and YKL in the corresponding positions which may also function as peroxisomal

targeting signals.

neighbor-joining method. The GenBank accession numbers were indicated in the

brackets. * indicates the branch from nematode-trapping fungi.

Supplementary Fig. 3 A neutral red staining of the hypha of the wild-type and

ΔAoHex1 strains. The two strains were cultured on PDA plates for 5 days. The hypha

of both strains were directly stained with neutral red staining solution (BBI Life

Sciences, Shanghai, China) for 10 minutes and rinsed twice with distilled water, 2

minutes each. The stained hypha were observed under a light microscope. A, the wild-

type strain. The cherry red spots indicate the vacuoles. B, the ΔAoHex1 strain.
