Retrograde Signaling through Cannabinoid ReceptorsThe bioactive agent from Cannabis, Δ9-tetrahydrocannabinol (THC),

activates two distinct cannabinoid (CB) receptors, CB1 and CB2, to elicit

a wide array of effects on mood, appetite, immunity, memory, and

pain perception. The receptors CB1 and CB2 are activated naturally by

endogenous CBs (endoCBs), including 2-arachidonoyl glycerol (2-AG).

While the CB2 receptor is abundant on leukocytes and has key roles in

immunoregulation and pain, the CB1 receptor predominates throughout

the central nervous system. Both are Gi-linked receptors, so that

activation blocks the action of adenylate cyclase, preventing signaling

through cyclic AMP. In the neuronal synapse, endoCBs like 2-AG act

in a retrograde fashion. That is, 2-AG is synthesized and secreted by

post-synaptic neurons and activate CB1 receptors at the presynaptic

axon terminal. Activation of CB1 then modulates signaling within the

presynapse, producing either a transient or prolonged reduction in the

release of neurotransmitters. For example, in the hippocampus, CB1

receptors are more abundant on inhibitory γ-aminobutyric acid (GABA)

terminals than on excitatory glutamate terminals. Activation of CB1 by

2-AG from post-synaptic terminals strongly decreases the release of the

inhibitory neurotransmitter GABA and less effectively reduces the release of the stimulatory glutamate. Prolonged inhibition of glutamate

signaling, as may occur with overproduction of endoCBs, may impair hippocampus-dependent learning and memory. Through its many

variable effects on several different types of neurotransmitters, CB1 signaling confers additional plasticity in neuronal signaling.

Cellular and Scrapie Forms of Prion ProteinPrion diseases include bovine spongiform encephalopathy

(BSE), scrapie of sheep and goats, chronic wasting disease

(CWD) of cervids, and human Creutzfeld Jakob disease

(CJD). Infectious transmission involves conversion of the

prion protein (PrP) from the cellular form PrPC to the disease-

associated scrapie prion, PrPSc. While PrPC is monomeric

and sensitive to proteinase K treatment in the presence

of detergents, PrPSc is prone to aggregation and partially

proteinase K resistant.

PrPC, encoded by the PRNP gene, undergoes

posttranslational modifications and is expressed as a

cell-surface glycosylated, GPI-anchored protein. The

C-terminal domain of PrPC forms a globular structure with

three α-helices (HI, H2, and H3) and two short anti-parallel

β-strands (S1 and S2), whereas the N-terminal domain is

highly flexible and lacks identifiable secondary structure.

H1 is an unusually hydrophilic α-helix located between

S1 and S2 with few contacts with the remainder of the

molecule, while H2 and H3 are hydrophobic, arranged anti-parallel and linked by a single disulfide bond. PrPC is composed of 42%

α-helix and 3% β-sheet while PrPSc contains 30% α-helix and 43% β-sheet. It is not clear whether PrPSc is produced from either mature PrPC

or an unfolded, intermediate isoform. Both conformations have potential to convert prion protein to a β-sheet-rich structure. Acting as a

seed, the β-sheet-rich conformation has a propensity to recruit additional β-sheet monomers to construct the irreversible, disease-related

isoform. As PrPSc monomers accumulate, the growing PrPSc polymers may break off into smaller fragments, which would enable further

dispersion of the pathogenic proteins and accelerate conversion of PrPC.
