Toxicity is the leading cause of drug failure, so

increasing the success rate of therapeutic

development requires more sensitive and rapid

methods to determine organ-specific damage.

The traditional tests to determine kidney toxicity

are blood urea nitrogen (BUN) and serum

creatinine (SCr), which are not optimal for early

detection or localization of kidney damage. As a

result, researchers have turned to recently

qualified nephrotoxicity biomarkers for more

predictive toxicity analysis. We have developed

the MILLIPLEX® MAP Rat Kidney Toxicity Magnetic

Bead Panels 1 and 2, containing a total of 14

kidney toxicity biomarkers.

We used these panels to efficiently profile

response to gentamicin, a known nephrotoxicant

in rats. By simply measuring biomarker

concentrations in urine, we detected kidney

toxicity (marked by increased Clusterin and

KIM-1 levels) in as few as three days (Figure 3).

Glucagon is the most important counter-

regulatory hormone to insulin in blood glucose

homeostasis. It stimulates gluconeogenesis and

glycogenolysis while inhibiting glycogenesis and

glycolysis mainly in the liver. Recently, glucagon

and its receptors have been actively pursued as

therapeutic targets for Type I diabetes. However,

quantifying glucagon in blood has been

challenging due to glucagon’s many

immunoreactive forms.

EMD Millipore’s new, highly specific glucagon

ELISA kit shows no cross-reactivity to

miniglucagon or glucagon 1–18 and < 5%

cross-reactivity to oxyntomodulin. This assay,

including sample extraction using our rapid

protocol (Figure 4B), can be completed within a

working day. In contrast to kits from competing

suppliers, results from our ELISA kit agree well

with published glucagon levels.

Figure 3A and 3B. 3 groups of 5 adult male Sprague Dawley® rats, each injected intraperitoneally daily with either water, low-dose (70 mg/kg) gentamicin or high-dose (140 mg/kg) gentamicin. Day 0 urine collection is after the 1st dose, and subsequent collections are at

days 3, 7 and 14. All nephrotoxicity biomarkers were analyzed, but we show data here only for Clusterin and KIM-1. Analyte concentrations in gentamicin treated animals spike at day 3 for both Clusterin and KIM-1, and decrease at days 7 and 14.

Handheld Magnetic Separator Block for 96-Well Flat Bottom Plates or 96-Well Conical Well Plates(Cat. No. 40-285)

The Handheld Magnetic Separator Block offers a low cost option

for biomagnetic separation. Magnets are positioned around the

bottom of the well to pull the beads to the side of the well. The

top magnetic frame is white polycarbonate, with a corrosion

resistant steel plate underneath, all mounted to a polypropylene

base. An adjustable clip system holds a wide variety of

microplates to the separator block, and an O-Ring on the base

plate facilitates gripping.
