OverviewThe API 2000™ LC/MS/MS System brings all of the power andperformance of triple quadrupole massspectrometry right to your benchtop.Other instruments claim to have abenchtop design — but use the entire bench. The API 2000 System leaves plenty ofroom for everything else in your lab.Its footprint measures a compact 45 cm X 100 cm.

In addition to the small footprint, the API 2000 LC/MS/MS System features robust ion sources fromApplied Biosystems/MDS SCIEX.These sources offer full articulationwith large viewing ports for easyoptimization. To prevent contamina-tion during switching, the sources are positively vented and include thepatented Curtain Gas™ interface tomaximize ion transfer to the massanalyzer and provide superior ruggedness and reliability. Sincesafety is always a concern, the ionsources are fully insulated so thatoperators are protected from hotsurfaces. Because time is so valuable, the ion sources for the API 2000System are truly “plug and play,” thus allowing for a quick change-over between the sources to keepoperations running smoothly withoutsacrificing productivity.

In general, triple quadrupole massspectrometers are very popular analytical instruments. They offermany key benefits to researchers who are interested in high throughputanalyte quantitation using a massdetector of high specificity coupled to a liquid chromatograph. Triplequadrupole MS/MS is one of the mostsensitive and specific technologiesavailable for small molecule quantita-tion. This technique is amenable to a broad range of compounds and inherently eliminates the need forextensive sample preparation. Thefragmentation capabilities of tandemmass spectrometry also provide critical information for structural elucidation of unknown compounds.

Some typical applications for triplequadrupoles include:

• Drug Metabolism andPharmacokinetics (DMPK)

• Environmental analysis

• Forensic toxicology

• Medical research

• Drug delivery

• Protein and peptide characterization

• Impurity and metabolite profiling

Key Features of the API 2000LC/MS/MS System• Small footprint

• LINAC™ collision cell for highthroughput Multiple ReactionMonitoring (MRM)

Proven PerformanceYou won’t have to sacrifice perform-ance for size. The API 2000™System’s proven engineering includesthe proven Curtain Gas™ interfaceand the LINAC™ collision cell. The latter allows for improved scanspeeds and increased sensitivity while preventing contamination and eliminating cross-talk.

The Curtain Gas interface employs a nitrogen stream flowing counter-current to the ion path. This gas flow drives solvents and buffers from the inlet, maximizing ion transfer to the analyzer, therebyproviding unmatched ruggedness and reliability. Efficient Ionization is achieved without heat. This eliminates the possibility of thermaldegradation of labile compounds.

The “Gold Standard” in Ionization PerformanceApplied Biosystems/MDS SCIEX offerstwo ion sources for the API 2000System to accommodate all of youranalytical demands:

Heated Nebulizer or Atmospheric Pressure ChemicalIonization (APCI) Source• Full gradient capabilities

• Computer controlled probetemperature and gas flow

• Optional Supercritical-FluidChromatography (SFC) compatibility

• Flow rates up to 2 mL per minute

• Allows ionization of neutral molecules

• Species are less susceptible toionization suppression

• Wide dynamic range

• LC “dead” volume is less of aconsideration

• Higher buffer concentrations are acceptable

TurboIonSpray™ Source• Permits flow rates from 2 µL to

1 mL per minute

• Exhibits excellent concentrationdependent response

• Ability to analyze high molecularweight compounds

• Very sensitive

• Wide range of accepted analytes

• Experiences none of the problemsassociated with thermal lability or volatility

Ease-of-UseNever before has an LC/MS/MS system been so user friendly. The APCI and TurboIonSpray sources can be completely inter-changed within minutes without tools. The following three stepsillustrate the ease with which thesources can be switched.

1. Flip the toggle bolts.

2. Angle and lift the source off.

3. Replace with the new source.

Rapid source change-over helps tominimize laboratory downtime andmakes the overall job of maintainingyour mass spectrometer much easier.

All temperature, gas and electricalconnections are fully integrated intothe source housing so there are noextra lines to attach. This seamlessconversion is completed when mag-netic connections automatically detectand indicate the hardware change tothe software. There is no need to“tell” the software what source youare using. The built-in syringe pumpand diverter valve simplify hardwareconfiguration and also contribute toconserving bench space.

Superior engineering allows for reliable flow rates as low as 2 µL perminute and up to 2 mL per minute. In addition, ion sources are safetyinterlocked — voltages are auto-matically set to zero when eithersource is removed.

All mass spectrometers requireoccasional cleaning and mainte-nance. Maintenance is simple withthe API 2000™ System. The systemcovers are easily removed with com-mon tools, and components such as the curtain plate, orifice, skimmer, and entrance quadrupole are readilyaccessible for cleaning.

The API 2000 System also featuresAnalyst™ software for acquisition, instrument control and data process-ing. Analyst software simplifies datareduction in the familiar Windows NT®

operating environment. Automatictuning and calibration ensureoptimum performance while rapiddata acquisition increases yourproductivity, allowing you to spendvaluable time exploring and inter-preting data instead of on system set-up. Analyst software furtherstreamlines your system by offeringsingle point control over a wideselection of LC pumps, autosamplersand liquid handling systems.

ConclusionsLaboratories of all sizes can benefitfrom the advantages of tandem massspectrometry with the API 2000LC/MS/MS System. This ruggedsystem gives you the high throughputand specificity that you need in an economical, benchtop platform.Productivity is improved even furtherwith the API 2000 System because of its rugged and easy to use ionsources. The Curtain Gas™ interfaceand LINAC™ collision cell areinstrumental in providing superiorperformance. Experience the benefitsof a triple quadrupole instrument with a smaller footprint and lessmaintenance than you would expect.
