Prinzip der Translation
• Synthese von Polypeptiden, wird von der mRNA gesteuert
• Die Zelle muss die Basenfolge eines mRNA-Moleküls in die Aminosäuresequenz eines Polypeptids übersetzen
• Die Translation ist somit der letzte Schritt in einem Prozess, bei dem anhand des genetische Codes aus Erbinformationen ein Eiweißmolekül hergestellt wird.
• Dieser letzte Schritt geschieht in lebenden Zellen an besonderen Strukturen, den Ribosomen

Der genetische Code
• Regel, nach der Dreiergruppen aufeinander folgender Nucleotide(Nucleobasen), Tripletts oder Codons genannt in Aminosäuren (AS) übersetzt werden, findet nur bei der Bindung der AS an tRNA statt.
• Nucleobasen: Adenin, Guanin, Cytosin, Uracil
• einige Codons stehen nicht für eine AS, sondern werden als STOPP-Codonbezeichnet, welche die Proteinsynthese beenden (Beispiel: UGA)
• AUG dient sowohl als Codon für Methionin als auch als Startsignal der Translation

Stufen der Peptidsynthese am Ribosom (Elongation)
1. Codonerkennung:Eine ankommende Aminoacyl-tRNAbindet an das an der A-Stelle befindliche Codon.
2. Peptidbindung:Zwischen der neuen AS und der wachsenden Polypeptidkette wird einePeptidbindung geknüpft.
3. Translokation:Die tRNA an der P-Stelle wird entlassen. Die tRNA an der A-Stelle wird zur P-Stelle verschoben. Bei diesem Vorgang bewegt sich das Ribosom um ein Codonin 5`→ 3`-Richtung vorwärts.

• Alle Proteine erhalten eine typische N-terminale Signal-Sequenz, die von einem speziellen Protein, dem signal recognition particle (SRP) erkannt wird.
• SRP bindet an ein Rezeptorprotein in der ER-Membran
• SRP wird freigesetzt, während sich das wachsende Polypeptid durch ein Kanalprotein in der ER-Membran schlängelt.
